摘 要:为了探究槲皮素对产肠毒素大肠杆菌K88诱导肠炎小鼠的治疗效果及对肠道菌群的影响,试验将50只小鼠随机分为空白对照(CON)组、模型(MO)组、槲皮素低剂量(QUE-L)组、槲皮素中剂量(QUE-M)组、槲皮素高剂量(QUE-H)组,每组10只。除CON组外,每组腹腔注射大肠杆菌菌液0.2 mL,QUE-L、QUE-M、QUE-H组在攻菌2 h后按照体重分别灌胃50,100,200 mg/kg的槲皮素药液0.2 mL,CON组和MO组给予等体积生理盐水,24 h后处死所有小鼠。采用H.E.染色观察小鼠十二指肠组织的病理损伤情况,采用ELISA试剂盒检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的质量浓度;基于16S rRNA基因测序分析小鼠盲肠内容物微生物α多样性及门、属水平的物种组成结构。结果表明:MO组十二指肠单层柱状上皮细胞出现坏死缺失,黏膜层有炎症细胞浸润;与MO组相比,槲皮素各剂量组十二指肠组织均得到不同程度的修复,其中QUE-H组修复效果最好,十二指肠单层柱状上皮细胞结构完整,仅伴随轻微的炎症细胞浸润。与CON组比,MO组TNF-α、IL-1β、IL-6质量浓度极显著升高(P<0.01),IL-10质量浓度极显著降低(P<0.01);与MO组相比,槲皮素各剂量组TNF-α、IL-1β和IL-6质量浓度显著或极显著降低(P<0.05或P<0.01),IL-10质量浓度极显著升高(P<0.01)。与CON组相比,MO组盲肠内容物微生物辛普森(Simpson)指数极显著降低(P<0.01),厚壁菌门相对丰度极显著降低(P<0.01),变形菌门相对丰度极显著升高(P<0.01),Muribaculaceae属和乳酸杆菌属相对丰度均极显著降低(P<0.01),埃希菌-志贺氏菌属相对丰度极显著升高(P<0.01)。与MO组相比,槲皮素各剂量组香浓(Shannon)指数和Simpson指数均极显著升高(P<0.01),QUE-H厚壁菌门相对丰度极显著升高(P<0.01),QUE-L、QUE-M、QUE-H组变形菌门相对丰度显著或极显著降低(P<0.05或P<0.01),QUE-M、QUE-H组乳酸杆菌相�In order to explore the therapeutic effects of quercetin on mice with enteritis induced by enterotoxigenic Escherichia coli(E.coli)K88 and its impact on intestinal flora,in the test,fifty mice were randomly divided into blank control(CON)group,model(MO)group,quercetin low-dose(QUE-L)group,quercetin medium-dose(QUE-M)group and quercetin high-dose(QUE-H)group,with 10 mice in each group.Except the CON group,each group was intraperitoneally injected with 0.2 mL of E.coli liquid.QUE-L,QUE-M and QUE-H groups were intragastrically injected with 0.2 mL of 50,100 and 200 mg/kg of quercetin liquid respectively according to body weight 2 h after bacterial challenge;CON group and MO group were given the same volume of normal saline;all mice were sacrificed after 24 h.Pathological lesion of duodenum of mice was observed by H.E.staining,and the mass concentrations of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)and interleukin-10(IL-10)in serum were detected by ELISA kit.Based on 16S rRNA gene sequencing,the microbialαdiversity and species composition structure at phylum and genus levels in cecum contents of mice were analyzed.The results showed that there was necrosis and loss of duodenal monolayer columnar epithelial cells and inflammatory cell infiltration in the mucosa in MO group.Compared with the MO group,the duodenal tissue of quercetin in each dose group was repaired to different degrees,among which the QUE-H group had the best repair effect,and the duodenal monolayer columnar epithelial cell structure was intact,accompanied only by slight inflammatory cell infiltration.Compared with the CON group,the mass concentrations of TNF-α,IL-1βand IL-6 in the MO group were significantly increased(P<0.01),and the mass concentration of IL-10 was significantly decreased(P<0.01).Compared with the MO group,the mass concentrations of TNF-α,IL-1βand IL-6 in quercetin dose groups decreased significantly or very significantly(P<0.05 or P<0.01),and the mass concentration of IL-10 increased very signifi
