泰乐菌素联合黄芩苷治疗耐药鸡毒支原体的药效学研究

摘  要:为了探究泰乐菌素联合黄芩苷对耐药鸡毒支原体的体内外药效学效果,试验采用倍比稀释法测定泰乐菌素和黄芩苷对鸡毒支原体的最低抑制浓度(MIC),通过逐渐增加泰乐菌素药物浓度诱导耐药,测定诱导后鸡毒支原体的MIC;使用棋盘稀释法测定泰乐菌素和黄芩苷的部分抑菌浓度指数(FICI);建立雏鸡鸡毒支原体感染模型,设置空白组、感染组、泰乐菌素组、黄芩苷组和联合用药组,研究联合给药对雏鸡感染耐药鸡毒支原体体内药效学效果,测定各组肺脏组织中鸡毒支原体含量及炎症因子的质量浓度;制作组织切片,观察给药后各组肺脏组织形态变化。结果表明:在体外泰乐菌素单用对鸡毒支原体的MIC为0.004μg/mL,黄芩苷单用对鸡毒支原体的MIC为31.25μg/mL。泰乐菌素诱导后,泰乐菌素单用对鸡毒支原体的MIC为2.56μg/mL,黄芩苷单用对鸡毒支原体的MIC为31.25μg/mL,两药联合使用的FICI为0.25,表现协同作用。在雏鸡感染模型中,泰乐菌素组、黄芩苷组和联合用药组雏鸡肺脏组织中鸡毒支原体含量均极显著降低(P<0.01),其中联合用药组极显著低于泰乐菌素组和黄芩苷组(P<0.01),抗菌效果优于单药治疗组。感染组、泰乐菌素组和黄芩苷组的白细胞介素-1(IL-1)、白细胞介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)质量浓度均显著高于联合用药组(P<0.05),而联合用药组与空白组差异不显著(P>0.05),联合用药组降低炎症反应优于单药治疗组。同时,联合用药可恢复肺组织结构,减少炎性细胞浸润。说明泰乐菌素与黄芩苷体外联合用药可对耐药鸡毒支原体表现协同作用,体内联合用药效果优于单一用药,可显著降低炎症因子水平,对肺脏组织损伤有良好的改善效果。In order to explore the in vitro and in vivo pharmacodynamic effects of tylosin combined with baicalin on drug-resistant Mycoplasmoides gallisepticum,in the test,the minimum inhibitory concentration(MIC)of tylosin and baicalin against Mycoplasmoides gallisepticum was determined by the doubling dilution method.Resistance was induced by gradually increasing tylosin drug concentrations,and the MIC of Mycoplasmoides gallisepticum after induction was determined.The partial inhibitory concentration index(FICI)of tylosin and baicalin was determined by checkerboard dilution method.A model of Mycoplasmoides gallisepticum infection in chicks was established.The blank group,infection group,tylosin group,baicalin group and combination drug group were set up,in order to study the in vivo pharmacodynamic effect of combination administration on the infection of drug-resistant Mycoplasmoides gallisepticum in chicks.The content of Mycoplasmoides gallisepticum and the mass concentrations of inflammatory factors in the lung tissues of each group were determined.Tissue sections were made to observe the morphological changes of lung tissues in each group after administration.The results showed that the MIC of tylosin alone against Mycoplasmoides gallisepticum was 0.004μg/mL,and the MIC of baicalin alone against Mycoplasmoides gallisepticum was 31.25μg/mL.After tylosin induction,the MIC of tylosin alone against Mycoplasmoides gallisepticum was 2.56μg/mL.The MIC of baicalin against Mycoplasmoides gallisepticum was 31.25μg/mL.The combination of the two drugs had a FICI of 0.25 and showed a synergistic effect.In the chick infection model,the content of Mycoplasmoides gallisepticum in the lung tissues of the tylosin group,baicalin group and combination group was significantly reduced(P<0.01),and the combination group was significantly lower than those of the tylosin group and baicalin group(P<0.01),and the antibacterial effect was better than that of the mono-drug group.The mass concentrations of interleukin-1(IL-1),interleukin-6(IL-6
