番茄顺式还原酮加双氧酶基因家族分析及功能鉴定

摘  要:顺式还原酮加双氧酶(acireductone dioxygenase, ARD)是Cupins蛋白家族的一员,在原核生物和真核生物中广泛存在,主要参与甲硫氨酸代谢途径的催化;另外,植物中甲硫氨酸代谢途径与乙烯和多胺等的合成密切相关,且乙烯和多胺参与了植物生长发育和逆境胁迫响应等多种生物学过程。植物中关于ARD的研究报道有限,尤其是园艺作物。番茄在人们的日常饮食结构中占有重要比例,也是植物研究领域常用的模式植物之一。本研究以番茄(Solanum lycopersicum)为研究对象,鉴定番茄基因组中包含的SlARD成员,并利用生物信息学分析手段、定量PCR技术和亚细胞定位等实验,对番茄SlARD基因家族进行分析及功能鉴定。结果表明,番茄基因组中共包含2个SlARD成员,分别命名为SlARD1和SlARD2,二者均定位于番茄第9号染色体,与拟南芥(Arabidopsis thalina)AtARD家族成员存在一定的系统发育和进化关系;SlARD1/2均只包含1个ARD结构域;番茄SlARD1/2在根、茎、叶中均有表达,能够响应水涝胁迫,定位于细胞质和细胞核。本研究还对结合SlARD1和SlARD2启动子的转录因子和SlARD1/2的互作蛋白网络进行了预测。上述实验结果能够为番茄SlARD的功能和分子机理研究提供理论参考和实验依据。The acireductone-dioxygenase(ARD)is a member of the Cupin protein family,widely present in prokaryotes and eukaryotes,and plays a critical role in catalyzing the methionine salvage pathway.This pathway in plants is closely related to the synthesis of ethylene,polyamines,and other compounds,which are involved in various biological processes such as plant growth,development,and responses to environmental stress.Researches on ARD in plants are limited,especially in horticultural crops.Tomatoes play an important role in human daily dietary structure and serve as one of the commonly used model plants in the field of plant research.This study identifies the members of the SlARD gene family within the tomato(Solanum lycopersicum)genome,investigates the SlARD gene family and characterizes their functions,utilizing bioinformatics analysis,quantitative Real-Time PCR techniques,and experimental approaches such as subcellular localization.The results indicate that there are two members of the SlARD gene family in the tomato genome,named SlARD1 and SlARD2,both located on the 9th chromosome of tomato.They show certain phylogenetic and evolutionary relationships with members of the AtARD family in Arabidopsis thalina. Both SlARD1 and SlARD2 contain a single ARD domain and are expressed in roots, stems, and leaves. They can respond to waterlogging stress, and are localized in the cytoplasm and nucleus. This study also predicted the transcription factors binding to the promoters of SlARD1 and SlARD2, as well as the protein-protein interaction network of SlARD1/2. The experimental results can provide theoretical references and experimental basis for the study of the function and molecular mecha-nisms of tomato SlARD.
