摘 要:核酸分析仪可用于检测茄子品种的简单序列重复(simple sequence repeats,SSR)的谱带迁移的多样性遗传标记。为验证核酸分析仪在茄子分子标记分析中的可行性和可靠性,本研究从实验室高通量测序获得的生物学数据和大量研究公布的茄子SSR序列信息中,筛选出100对SSR引物序列,利用核酸分析仪对茄子杂交种‘闽茄6号’及其亲本进行检测。从合成的100对引物中,获得10对具有多态性的SSR引物,不同引物的核酸多态性检测图在峰的数量和荧光强度上存在明显差异。结果表明核酸分析仪可用于图谱扩增,且扩增出的核酸多态性检测图与基因型一一对应。其中有1对引物ESSR67可在杂交种‘闽茄6号’DNA上同时获得父母本的特征峰,表现为双亲互补型。利用引物ESSR67通过比对茄子杂交种‘闽茄6号’和育种亲本的DNA样本的多态性峰型,可鉴定‘闽茄6号’茄子杂交种种子的真实性及纯度,并通过田间植物生物学性状观察相互验证。核酸分析仪因其超高灵敏度、高分辨率、样品需求量少、安全、操作简单等优点,可用于‘闽茄6号’杂交种纯度的鉴定研究,将是杂交茄子种子真实性和纯度快速鉴定的有效手段。Nucleic acid analyzer can be used to detect the diversity genetic markers of band migration in simple sequence repeats(SSR)of eggplant cultivars.In order to test the feasibility and reliability of the nucleic acid analyzer for molecular marker genotyping in eggplant,the nucleic acid analyzer analysis on'Minqie 6'and their parents were conducted.100 pairs of SSR primer sequences were selected from biological data obtained from high-throughput sequencing in the laboratory and from the sequence information of eggplant SSR published in many studies.10 pairs of SSR primers with polymorphism were obtained from the 100 pairs of primers synthesized.The nucleic acid polymorphism detection plots of different primers showed obvious differences in the number of peaks and the fluorescence intensity.The results of sequencing showed that nucleic acid analyzer amplification map,nucleic acid polymorphism detection map and genotype one-to-one correspondence.Among them,one pair of primers,ESSR67,could simultaneously obtain the characteristic peak of parent book on the hybrid'Minqie6'DNA,showing the complementary type of parent.The authenticity and purity of the seeds of the eggplant hybrid'Minqie 6'and the DNA samples of the breeding parents can be identified by using the primer ESSR67.The results could be identified and mutually verified by the identification of morphological traits in the field.The nucleic acid analyzer,which has the advantages of high sensitivity,high resolution,low sample demand,safety and simple operation,can be used for the identification of the purity of the hybrid species of'Minqie 6',which will be an effective means for the rapid identification of the authenticity and purity of the hybrid eggplant seeds.
