摘  要:酸枣具有很高的经济价值和营养价值,是黄河三角洲地区盐碱地开发的重要树种。以盆栽酸枣幼苗为试材,用200 mmol·L^(-1)NaCl溶液处理6 h、12 h、24 h、36 h,以超纯水为对照,检测叶片相对电导率、丙二醛含量、SOD和POD活性等生理指标的变化,并将处理6 h的试材进行转录组测序和分析,初步探究盐胁迫对酸枣幼苗叶片转录水平的影响。结果表明:盐胁迫下,酸枣幼苗叶片相对电导率、丙二醛含量呈现增加趋势,POD活性呈现上升趋势,SOD呈先上升后下降趋势;转录组测序筛选获得2974个DEGs,GO富集集中在膜、细胞质膜、信号转导等,KEGG富集集中在植物激素信号转导、淀粉和蔗糖代谢等通路;DEGs中有转录因子bZIP家族成员2个、WRKY家族成员13个、MYB家族成员18个、NAC家族成员6个、bHLH家族成员9个。研究结果可为探究酸枣响应盐胁迫的分子机制提供参考。Jujube has high economic value and nutritional value,and is an important tree species for the development of saline and alkaline land in the Yellow River Delta region.Potted jujube seedlings were treated with 200 mmol·L-1 NaCl solution for 6 h,12 h,24 h and 36 h,and clean water was used as the control,to detect the changes of physiological indexes,such as relative conductivity,malondialdehyde content,SOD and POD activities of leaves,etc.Transcriptome sequencing and analysis were also performed on the control and the 6 h-treated test material,to preliminarily investigate the effects of salt stress on the transcript levels of jujube seedlings.The results showed that:under salt stress,the relative conductivity and malondialdehyde content of jujube seedlings showed an increasing trend,POD activity showed an increasing trend,and SOD showed a first increasing and then decreasing trend;transcriptome sequencing screening obtained 2974 DEGs,GO enrichment focused on the membrane,cytoplasmic membrane,signaling,etc.,and KEGG enrichment focused on the phytohormone signaling,starch and sucrose metabolism and other pathways;There are 2 members of transcription factor bZIP family,13 members of WRKY family,18 members of MYB family,6 members of NAC family and 9 members of bHLH family in DEGs.The results of the study may provide a reference for exploring the molecular mechanism of jujube in response to salt stress.
