摘 要:硒代半胱氨酸(Sec)作为硒在生命体系中的主要存在形式,其含量和氧化还原状态的变化与诸多生理过程和疾病相关。文章以荧光素衍生物为母体合成了一种在弱酸条件下能够特异性响应硒代半胱氨酸的荧光探针。该探针能够实现纯水溶剂中对微量硒代半胱氨酸的快速响应,不会受常见金属离子、氨基酸以及生物硫醇的影响。该探针在pH=5的缓冲溶液中对硒代半胱氨酸的检测表现出较高的灵敏度,响应前后荧光恢复约163倍,为水环境中Sec的选择性检测提供了一种新的方法和思路。Selenocysteine(Sec)is the main form of selenium in living systems,and its content and oxidation-reduction state changes are related to many physiological processes and diseases.A fluorescent probe based on fluorescein derivative that can specifically respond to selenocysteine in weak acid conditions was synthesized in this study.The probe can achieve a fast response to trace amounts of selenocysteine in pure water solvent,and common metal ions,amino acids or biothiols did not interfere with the detection.The probe exhibits high sensitivity for the detection of selenocysteine in a buffer solution at pH=5,with a fluorescence intensity enhancement of about 163 times before and after the response,providing a new method for the selective detection of Sec in water environments.
