加味少腹逐瘀汤对子宫内膜异位症痛经IL-6/GP130通路的调控研究

摘　　要：目的基于白介素6(Interleukin-6,IL-6)/糖蛋白130(Glycoprotein 130,GP130)通路探讨加味少腹逐瘀汤对寒湿瘀结证子宫内膜异位症痛经小鼠腹腔液辅助性T淋巴细胞1(T-helper 1,Th1)/辅助性T淋巴细胞2(T-helper 2,Th2)的影响。方法采用腹腔注射法和冰水浴法构建寒湿瘀结证子宫内膜异位症痛经模型。采用HE、Masson染色比较病理变化;Western Blot测定蛋白表达;流式细胞学检测腹腔液中Th1、Th2细胞;Elisa测定白介素4(Interleukin-4,IL-4)、白介素10(Interleukin-10,IL-10)、白介素12(Interleukin-12,IL-12)、INF-γ(interferon-γ,INF-γ)。结果药物干预后,各治疗组疼痛潜伏期延长、扭体次数减少、盆腔粘连减轻,异位病灶总体积减少(P<0.05);病理显示药物干预后炎症改善,胶原沉积减轻;药物干预后异位组织IL-6、IL-6R、GP130蛋白表达降低;腹腔液中Th1细胞比例增高,Th2细胞比例下降;外周血IL-4、IL-10降低、外周血IL-12、INF-γ升高。结论加味少腹逐瘀汤可以改善寒湿瘀结证子宫内膜异位症小鼠腹腔液Th1/Th2平衡及调节IL-6/GP130信号通路,进而减轻模型小鼠腹腔炎症状态。Objective This study aims to explore the effects of Jiawei Shaofu Zhuyu Decoction on the symptoms of dysmenorrhea in mice with cold-dampness stasis and endometriosis,as well as its impact on the balance of T-helper 1(Th1)/T-helper 2(Th2)cells in the peritoneal fluid,based on the interleukin-6(IL-6)/Glycoprotein 130(GP130)signaling pathway.Methods The model of dys-menorrhea in mice with cold-dampness stasis and endometriosis was established using abdominal injection and ice water bath method.Hematoxylin-eosin(HE)staining and Masson trichrome staining were used to compare the pathological changes.Western blot was used to determine the protein expression levels of IL-6,IL-6R,and GP130.Flow cytometry was used to detect the number of Th1 and Th2 cells.ELISA was used to measure the levels of interleukin-4(IL-4),interleukin-10(IL-10),interleukin-12(IL-12),and interferon-γ(INF-γ)in the peripheral blood.Results After drug intervention,compared with the model group,the treatment groups showed prolonged pain latency,reduced twisting frequency,alleviated pelvic adhesion,decreased overall volume of ectopic lesions,and significant results in the high-dose herbal medicine group and positive drug group(P<0.05).Pathological examination showed improved inflammatory response and reduced collagen fiber deposition after drug intervention.The expression of IL-6,IL-6R,and GP130 proteins in ectopic tissue decreased after drug intervention.The proportion of Th1 cells in the peritoneal fluid increased,while the proportion of Th2 cells decreased.The levels of IL-4 and IL-10 in the peripheral blood decreased,while the level of INF-γand IL-12 increased.Conclusion Jiawei Shaofu Zhuyu Decoction can improve the Th1/Th2 balance in the peritoneal fluid of mice with cold-dampness stasis and endometriosis,may be related to the regulation of the IL-6/GP130 signaling pathway,reduce the inflammatory state of the abdominal cavity and clinical symptoms in model mice.
