猫白血病病毒山西毒株的遗传变异特征分析

摘  要:为了阐明山西省猫白血病病毒(FeLV)相似性及遗传变异特征,试验先对疑似感染FeLV的20份猫外周血提取病毒RNA,然后PCR扩增FeLV山西株env和pol基因,再采用相似性比对、系统进化树构建等方法对FeLV山西株env和pol基因、氨基酸变异水平和遗传进化特征进行研究。结果表明:共获得3株FeLV山西株,FeLV山西株间env基因相似性为97.7%~99.2%,pol基因相似性为97.0%~97.3%;在GenBank已公布的参考株中,FeLV山西株与我国黑龙江省毒株(MW839565.1)的相似性最高,其中env基因为97.5%~99.2%,pol基因为97.0%~97.4%;3株FeLV山西株均属于内源性猫白血病病毒(enFeLV),且env与pol蛋白均存在不同程度的氨基酸突变。3株FeLV山西株均与enFeLV处于同一个大的分支中,且分属于两个不同的小分支。说明env、pol基因均可用于FeLV相似性与遗传进化特征分析,3株FeLV山西株均属于enFeLV,与黑龙江省分离毒株的相似性最高,基因变异水平较低,但仍存在一定水平的氨基酸突变。To elucidate the homology and genetic variation characteristics of feline leukemia virus(FeLV)strains in Shanxi Province,this study first amplified the env and pol genes of the FeLV Shanxi strain.Subsequently,methods such as similarity alignment and phylogenetic tree construction were used to analyze the env and pol genes,amino acid variation levels and genetic characteristics of the FeLV Shanxi strain.Results showed that the homology of env and pol gene among FeLV Shanxi strains were 97.7%-99.2% and 97.0%-97.3%,respectively.Among the reference strains published in GenBank,the FeLV Shanxi strains exhibited the highest homology with the Heilongjiang strains(MW839565.1),with the env gene concordance rate of 97.5%-99.2%and pol gene 97.0%-97.4%.All three FeLV Shanxi strains belonged to endogenous feline leukemia virus(enFeLV),and amino acid site mutations were observed in both env and pol proteins.Phylogenetic analysis results of both env and pol genes revealed that the three FeLV Shanxi strains clustered within the same major clade as enFeLV but were distributed across two distinct subclades.In conclusion,the present study indicated that both env and pol genes could be used to analyze the homology and genetic development characteristics of FeLV.The FeLV Shanxi strains belonged to enFeLV,and exhibited low level of gene variation,with the highest level of homology to domestic isolates,although some amino acid site mutations were still present.
