改良生肌膏对创伤愈合过程中基质金属蛋白酶及其抑制剂含量的影响

摘  要:为探究改良生肌膏对SD大鼠皮肤创伤愈合过程中创面组织中MMP-1、MMP-2、MMP-9和TIMP-1、TIMP-2含量的影响,试验选取体重为(200±20)g的健康雄性SD大鼠60只,适应性喂养1周后制备皮肤缺损模型,将造模后SD大鼠随机分为给药组(皮肤缺损处涂抹改良生肌膏,1次/d)和对照组(不处理),每组各30只。在愈合过程中,测定各组平均创面愈合时间和愈合后的瘢痕面积,在试验第1,3,7,10,14天,每组各随机选取大鼠6只,用手术剪剪取大鼠创面中心至创缘正常皮肤间的组织,采用ELISA方法检测创面组织中MMP-1、MMP-2、MMP-9及TIMP-1、TIMP-2的含量。结果表明:给药组的平均创面愈合时间极显著短于对照组(P<0.01),愈后瘢痕面积极显著小于对照组(P<0.01)。造模后第1,3,7,10,14天,给药组MMP-9、TIMP-1含量极显著高于对照组(P<0.01);造模后第3天,给药组MMP-1含量显著高于对照组(P<0.05),MMP-9、TIMP-1、TIMP-2含量极显著高于对照组(P<0.01);造模后第7天,给药组TIMP-1、TIMP-2含量极显著高于对照组(P<0.01);造模后第10天,给药组MMP-1、MMP-9、TIMP-1、TIMP-2含量极显著高于对照组(P<0.01);造模后第14天,给药组MMP-1、MMP-9、MMP-2、TIMP-2含量极显著高于对照组(P<0.01)。说明改良生肌膏能缩短SD大鼠皮肤创口愈合时间,减小愈后瘢痕面积,影响MMPs和TIMPs含量。In order to explore the effects of modified Shengji ointment on the contents of MMP-1,MMP-2,MMP-9,TIMP-1 and TIMP-2 in wound tissues during the healing process of skin wound in SD rats,in the experiment,a total of 60 healthy male SD rats weighing(200+20)g were selected.After 1 week of adaptive feeding,a skin defect model was prepared.After modeling,they were randomly divided into drug group(application of modified Shengji ointment on the skin)and control group(no treatment),with 30 animals in each group.During the healing process,the average wound healing time and post-healing scar area of each group were measured.On the 1st,3rd,7th,10th and 14th days of the experiment,6 rats were randomly selected from each group,and the tissues between the center of the wound and the normal skin of the wound margin were cut by surgical scissors,and the contents of MMP-1,MMP-2,MMP-9,TIMP-1 and TIMP-2 in the wound tissues.were detected by ELISA.The results showed that the average wound healing time of the administration group was significantly lower than that of the control group(P<0.01),and the positive scar surface after healing was significantly lower than that of the control group(P<0.01).On the lst,3rd,7th,10th and 14th days after modeling,the contents of MMP-9 and TIMP-1 in the administration group were significantly higher than those in the control group(P<0.01).On the third day after modeling,the content of MMP-1 in the administration group was significantly higher than that in the control group(P<0.05),and the contents of MMP-9,TIMP-1 and TIMP-2 were significantly higher than those in the control group(P<0.01).On the 7th day after modeling,the contents of TIMP-1 and TIMP-2 in the administration group were significantly higher than those in the control group(P<0.O1).On the 1Oth day after modeling,the contents of MMP-1,MMP-9,TIMP-1 and TIMP-2 in the administration group were significantly higher than those in the control group(P<0.01).On the 14th day after modeling,the contents of MMP-1,MMP-9,MMP-2 and TIMP-2 in the admini
