摘  要:为了制备鸭疫里默氏杆菌(Riemerella anatipestifer,RA)菌影并探究其与CpG质粒联用的口服免疫效果,试验用NaOH将RAGD1904-2菌株制备成RA菌影,用扫描电镜检测菌影形态,将制备成功的RA菌影与CpG质粒联合应用。将80只1日龄的健康樱桃谷鸭随机分为对照组、高剂量RA菌影组(接种菌影1×10^(10) cfu)、低剂量RA菌影组(接种菌影1×10^(9 )cfu)、CpG+低剂量RA菌影组(CpG质粒50μg+接种菌影1×10^(9 )cfu),每组20只,分别在5日龄及14日龄进行口服免疫,每日1次,连续3 d;于28日龄采集各组鸭外周血并检测血清IgG、IgA、IL-2、IL-6含量;以1×10^(7.39) cfu(半数致死量)的RAGD1904-2菌株进行攻毒,攻毒后连续观察7 d,统计发病率及死亡率。结果表明:各免疫组血清IgG、IgA、IL-2、IL-6含量均极显著高于对照组(P<0.01),且CpG+低剂量RA菌影组血清IgG、IgA含量均极显著高于高剂量RA菌影组和低剂量RA菌影组(P<0.01);CpG+低剂量RA菌影组IL-2含量显著高于低剂量RA菌影组(P<0.05),IL-6含量极显著高于低剂量RA菌影组(P<0.01);高剂量RA菌影组血清IgG、IgA含量显著高于低剂量RA菌影组(P<0.05)。高剂量RA菌影组和低剂量RA菌影组发病率均高于50%,CpG+低剂量RA菌影组发病率为35%,死亡率仅为20%。说明RA菌影与CpG质粒的联合应用可以有效降低RA感染的发病率和死亡率,提高血清IgG、IgA、IL-2、IL-6含量。In order to prepare the Riemerella anatipestifer(RA)ghosts and explore the oral immune effect of its combination with CpG plasmid,in the test,RA ghosts were prepared by chemical method from cryopreserved RACD1904-2 strain,and the morphology of the ghosts was detected by electron microscope.The prepared microbial ghosts were combined with the CpG plasmid.A total of 801-day-old healthy Cherry Valley ducks were randomly divided into control group,high-dose RA ghosts group(1×10^(10)cfu),low-dose RA ghosts group(1×10^(9 )cfu)and CpG plasmid(50μg/each)+low-dose RA ghosts group(1×10^(9 )cfu),with 20 ducks in each group.Oral immunization was performed at 5 and 14 days of age,respectively;1 time a day,for 3 days.At 28 days of age,the peripheral blood of each group was collected and the serum levels of IgG,IgA,IL-2 and IL-6 were detected.1x107.39 cfu(half lethal dose)of RACD1904-2 bacteria were challenged;ater challenge it was observed continuously for 7 days and morbidity and mortality were counted.The results showed that the contents of IgG,IgA,IL-2 and IL-6 in serum of each immunization group were significantly higher than those in the control group(P<0.01).The contents of IgG and IgA in serum of the CpG plasmid+low-dose RA ghosts group were significantly higher than those of the high-dose and low-dose RA ghosts groups(P<0.01).The contents of IL-2 in the peripheral blood of the CpG plasmid+low-dose RA ghosts group was significantly higher than that in the low-dose RA ghosts group(P<0.05).The contents of IL-6 in serum of the CpG plasmid+low-dose RA ghosts group was significantly higher than that of the low-dose RA ghosts group(P<0.01).The contents of IgG and IgA in serum of the high-dose RA group were significantly higher than those in the low-dose group(P<0.05).The incidence rate of both high-dose and low-dose RA ghosts groups was higher than 50%,and the incidence rate of CpG plasmid+low-dose RA ghosts group was 35%,with the mortality rate of only 20%.The results suggested that the combination of RA and CpG plasmid
