摘　　要：目的观察脂肪间充质干细胞(adipose-derived stem cells,ADSCs)治疗糖尿病皮肤溃疡(diabetic cuta⁃neous ulcers,DCU)的作用。方法将ADSCs与人皮肤微血管内皮细胞(human dermal microvascular endothe⁃lial cells,HDMVECs)于高糖培养基中共培养,观察不同血糖浓度下HDMVECs的增殖、凋亡、迁移及成管能力。结果高糖促进HDMVECs凋亡,抑制其增殖及血管再生;ADSCs共培养HDMVECs后,恢复高糖下HDMVECs的增殖及血管再生能力。结论ADSCs在体外高血糖环境下共培养HDMVECs可以促进皮肤血管内皮的增殖能力和成管化,促进微血管形成改善微循环从而加速糖尿病皮肤溃疡愈合。Objective To observe the effect of adipose mesenchymal stem cells(adipose-derived stem cells,ADSCs)in the treatment of dia⁃betic skin ulcers(diabetic cutaneous ulcers,DCU).Method ADSCs were cocultured with human skin microvascular endothelial cells(human dermal microvascular endothelial cells,HDMVECs)in high glucose medium to observe the proliferation,apoptosis,migration and tube formation capacity of HDMVECs at different blood glucose concentrations.Result High glucose promoted HDMVECs apoptosis and inhibited its proliferation and vascular regeneration;After co-culture of ADSCs for HDMVECs,the proliferation and vascular regeneration capacity of HDMVECs under high glucose were restored.Conclusion Co-culture of HDMVECs by ADSCs in vitro hyperglycemia can pro⁃mote the value-added ability and tube formation of skin vascular endothelium,promote microvessel formation to improve microcirculation and thus accelerate the healing of diabetic skin ulcer.
