罗非鱼饲料绿原酸发酵配比及时长的筛选研究

摘  要:在罗非鱼商品饲料中添加枯草芽孢杆菌菌液,利用枯草芽孢杆菌发酵饲料,测定不同配比及时长下发酵饲料中绿原酸的含量。设置5个试验组,分别为A、B、C、D和E组。对应枯草芽孢杆菌液体积(mL)和商品饲料质量(g)的比例为1∶2、1∶3、1∶4、1∶5,不添加枯草芽孢杆菌液的饲料为E组,饲料发酵时长分别为6,12,18,24,30,36,42和48 h。采用高效液相色谱法测定饲料中绿原酸的含量。结果表明,42 h时,各组饲料中绿原酸含量均最高(P<0.05),6 h时均最低。C组和D组绿原酸含量变化趋势基本一致,且各发酵时长绿原酸含量均显著高于A组和B组(P<0.05)。A、B、C、D组绿原酸含量随发酵时间增加总体呈“升→降→升→降”的趋势。双因素分析结果表明,配比浓度(因素A)、发酵时长(因素B)及该双因素交互作用(因素A×因素B),均对绿原酸产量的合成有极显著影响(P<0.01);根据各因素F值大小,各因素影响的主次顺序为:因素A、因素B、因素A×因素B。指出,枯草芽孢杆菌液和饲料的最佳配比为1∶5,最佳的发酵时长为42 h。Bacillus subtilis bacterial liquid was added into tilapia commercial feed,and then the the content of chlorogenic acid was dected at different ratios and durations after being fermented in the feed.There were five experimental groups set up,and the ratio of the volume of Bacillus subtilis liquid(mL)to the mass of commercial feed(g)were 1∶2(Group A),1∶3(Group B),1∶4(Group C),1∶5(Group D).The feed was Group E without Bacillus subtilis liquid,and the fermentation time of the feed were 6,12,18,24,30,36,42,and 48 hours,respectively.Finally high performance liquid chromatography was used to determine the content of chlorogenic acid in the feed.The results showed that the content of chlorogenic acid in all groups of feed was highest at 42 hours(P<0.05),and lowest at 6 hours.The trend of chlorogenic acid content changes in Group C and Group D were basically consistent.And the chlorogenic acid content in Group C and Group D were significantly higher than those in Group A and Group B at all fermentation times(P<0.05).With fermentation time going by,the content of chlorogenic acid in groups A,B,C,and D showed an overall trend of“increase→decrease→increase→decrease”.The results of the two factor analysis showed that the single factor of ratio concentration(A),fermentation duration(B),and the interaction of these two factors(A×B)all had significant impacts on the synthesis of chlorogenic acid yield(P<0.01).According to the magnitude of each factor’s F-value,the order of priority for each factor was:A,B,A×B.The optimal ratio of Bacillus subtilis addition to feed was 1∶5,and the optimum fermentation duration was 42 hours.
