摘　　要：去泛素化酶含OTU结构域蛋白3(OTU domain-containing protein 3,OTUD3)是非常重要的去泛素化酶,在多种疾病模型中存在明显的表达差异。在不同细胞中,OTUD3参与调控凋亡、炎症反应、细胞周期、增殖和侵袭等生物学功能。OTUD3通过调节多种关键底物蛋白的去泛素化,在先天性抗病毒免疫、神经发育、神经系统退行性疾病、肿瘤等生理和病理过程中发挥重要作用。OTUD3在乳腺癌、食管癌、结肠癌、甲状腺乳头状癌中发挥抑癌作用,但在肝癌和肺癌中发挥促癌作用。作为恶性肿瘤的生物标志物,OTUD3在预测、诊断和判断预后方面起重要作用。OTUD3在大部分疾病中的相关分子机制研究仍不够深入,研究OTUD3的调控机制对于深入了解其在免疫和疾病中的作用至关重要,以OTUD3为靶标的药物研发将是今后的研究重点。OTU domain-containing protein 3(OTUD3)is a crucial deubiquitinase that exhibits significant expression differences across various disease models.OTUD3 plays a role in regulating biological functions such as apoptosis,inflammatory responses,cell cycle,proliferation,and invasion in different cell types.By deubiquitinating key substrate proteins,OTUD3 is involved in essential physiological and pathological processes,including innate antiviral immunity,neural development,neurodegenerative diseases,and cancer.OTUD3 exhibits tumor-suppressive effects in breast cancer,esophageal cancer,colon cancer,and papillary thyroid cancer,but acts as an oncogenic in liver and lung cancers.OTUD3 serves as a biomarker in predicting diagnosing,and assessing prognosis for certain malignancies.Despite its potential,the molecular mechanisms of OTUD3 in many diseases are still not well-understood,and exploring OTUD3’s regulatory mechanisms is essential for comprehending its roles in immunity and disease.Future research will focus on developing OTUD3-targeted therapies.
