PVP-Lys亲水/促纤溶改性聚砜基血液透析膜的制备及性能

摘　　要：针对透析膜材料血液相容性需要进一步提高的问题,采用非溶剂诱导相转化法制备聚砜膜,合成单端固定赖氨酸的聚乙烯吡咯烷酮(PVP-Lys),以PVP-Lys为间隔臂将赖氨酸修饰到聚砜基膜表面,开发了一种具有纤溶活性的抗血栓血液透析膜,通过扫描电子显微镜、X射线光电子能谱和水接触角测试等方法对膜进行表征,研究修饰不同含量赖氨酸的聚砜血液透析膜的性能。结果表明:成功制备了单端固定赖氨酸的PVP-Lys;PVP-Lys的加入使聚砜膜的表面结构发生变化,分离层的厚度增加;当PVP-Lys溶液质量分数为1.5%时,改性膜的BSA截留率为99.1%;与聚砜纯膜相比,亲水性提高了32%,抗蛋白质吸附率提升了67%,具有更高的抗血小板、红细胞粘附能力和更低的溶血率,改善了透析膜的血液相容性。Aiming at the issue that hemocompatibility of dialysis membrane materials needs to be further improved,polysulfone membranes were prepared by the nonsolvent induced phase separation method,polyvinylpyrrolidone with lysine immobilized at the single end(PVP-Lys)was synthesized,and lysine was immobilized onto the surface of the polysulfone membrane with polyvinylpyrrolidone as spacer to obtain an antithrombotic hemodialysis membrane with fibrinolytic activity.The membrane was characterized by scanning electron microscopy,X-ray photoelectron spectroscopy and water contact angle test to investigate the performance of polysulfone hemodialysis membranes modified with different lysine contents.The results show that the polyvinylpyrrolidone with single-end immobilized lysine is successfully prepared;the addition of PVP-Lys changes the surface structure of the polysulfone membrane and increases the thickness of the separating layer.The retention rate of BSA is 99.1%at a concentration of 1.5%(mass fraction)of PVP-Lys solution,which improves the hydrophilicity by 32%and the resistance to protein adsorption by 67%compared with polysulfone pure membrane,and possesses higher anti-platelet,red blood cell adhesion ability and lower hemolysis rate,and improves the hemocompatibility of dialysis membrane.
