摘 要:皮肤作为人体最大的器官,保护身体免受外界环境的影响.然而,随着各种因素的影响使皮肤逐渐衰老,甚至引发皮肤癌.胶原蛋白的降解是皮肤衰老的主要原因.胶原蛋白填充剂不仅可以直接补充胶原,还可以刺激成纤维细胞产生新的胶原.然而,目前的胶原填充剂存在不稳定、易降解的问题.交联胶原填充剂的稳定性提高,但交联剂可能引发细胞毒性和组织钙化的风险,因此在临床应用方面受限.本研究开发了一种新型的、长效的交联牦牛胶原蛋白填充剂用于皮肤年轻化.低浓度的交联剂可以高效交联牦牛胶原蛋白,形成较粗、网状结构均一的胶原纤维.交联后形成具有良好力学强度、长效性的牦牛胶原蛋白填充剂.交联牦牛胶原蛋白具有显著的热稳定性和持久性.此外,交联牦牛胶原填充剂是一种无细胞毒性、高生物活性的植入材料,可有效促进细胞的增殖和黏附.交联牦牛胶原填充剂植入小鼠皮下后能有效促进胶原蛋白再生,并具有良好的抗钙化潜能.交联牦牛胶原填充剂能有效促进胶原蛋白再生,并在皮肤年轻化治疗方面具有显著的潜力.As the largest organ of the human body,the skin serves as a protective barrier against the external environment.However,under the influence of various factors,the skin undergoes gradual aging and may even predispose individuals to skin cancer.The degradation of collagen stands as the primary culprit behind skin aging.However,currently collagen implants were susceptible to degradation.The durability of crosslinked collagen implants was enhanced;however,the utilization of crosslinking agents may entail the risk of cytotoxicity and tissue calcification,thereby constraining their clinical utilization.In this study,a novel and durable crosslinked yak collagen implant was developed for skin rejuvenation.Employing a low concentration of crosslinking agent efficiently facilitated the formation of collagen fibers with a thicker,and more uniform network structure.Post-crosslinking,the resultant yak collagen implant exhibited commendable mechanical strength and long-lasting.Furthermore,crosslinked yak collagen demonstrated remarkable thermal stability and durability.Furthermore,the crosslinked yak collagen implant effectively promoted cell proliferation and adhesion.Following subcutaneous implantation in mice,the crosslinked yak collagen implant effectively stimulated the regeneration of collagen and demonstrated commendable potential in resisting calcification.The crosslinked yak collagen implant effectively fosters collagen regeneration and holds considerable potential in the realm of skin rejuvenation treatment.
