摘 要:目的借助网络药理学探讨紫苏叶治疗登革病毒(DENV)的活性成分、作用靶点及潜在作用机制。方法借助TCMSP检索紫苏叶有效成分,整合PubChem、SwissTargetPrediction和UniProt数据库预测潜在靶点。应用OMIM和GeneCards数据库收集与DENV相关的靶点,构建“中药-成分-靶点-疾病”网络,筛选出核心活性成分。然后取紫苏叶与DENV的交集靶点,应用STRING数据库和Cytoscape软件构建靶点蛋白相互作用(PPI)网络,使用CytoHubba插件筛选关键基因并进行基因本体论(GO)分析和京都基因与基因组百科全书(KEGG)信号通路富集分析。结果共得到13种紫苏叶活性成分,515个活性成分作用靶点,1346个DENV相关靶点,98个交集靶点。紫苏叶的关键活性成分包括γ-谷甾醇和木犀草素等;关键基因包含AKT丝氨酸/苏氨酸激酶1、肿瘤坏死因子(TNF)、白细胞介素(IL)-6、丝裂原活化蛋白激酶3、半胱氨酸蛋白酶3、丝裂原活化蛋白激酶14、丝裂原活化蛋白激酶1、前列腺素H合成酶2、酪氨酸激酶1和FYN原癌基因,Src家族酪氨酸激酶基因。KEGG分析显示,这些关键基因主要涉及TNF信号通路和IL-17信号通路等。结论本研究初步揭示紫苏叶抗DENV作用机制,为后续研究提供理论支撑。Objective To explore the active components,action targets and potential mechanism of action of Perillae Folium for the treatment of Dengue virus(DENV)with the aid of network pharmacology.Methods The active ingredients of Perillae Folium were searched with the aid of TCMSP.The PubChem,SwissTargetPrediction and UniProt databases were integrated to predict potential targets.The OMIM and GeneCards databases were used to collect DENV-related targets to construct a"traditional Chinese medicine-ingredient-target-disease"network and screen out the core active ingredients.The intersecting targets of Perillae Folium and DENV were subsequently taken.The STRING database and Cytoscape software were applied to construct the target protein-protein interaction(PPI)network.The key genes were screened and subjected to gene ontology analysis and Kyoto encyclopedia of genes and genomes signaling pathway enrichment analysis using the CytoHubba plug-in.Results A total of 13 active ingredients of Perillae Folium,515 active ingredient action targets,1346 DENV related targets,and 98 intersecting targets were obtained.The key active ingredients of Perillae Folium includedγ-sitosterol and luteolin,etc.The key genes included AKT serine/threonine kinase 1,tumor necrosis factor(TNF),interleukin(IL)-6,mitogen-activated protein kinase 3,caspase 3,mitogen-activated protein kinase 14,mitogen-activated protein kinase 1,prostaglandin-endoperoxide synthase 2,janus kinase 1,and the FYN proto-oncogene,Src family tyrosine kinase.KEGG analysis showed that these key genes were mainly involved in the TNF signaling pathway and IL-17 signaling pathway.Conclusion This study preliminarily reveals the anti-DENV mechanism of action of Perillae Folium,and provides theoretical support for subsequent studies.
