不同给药途径裘氏内异方对子宫腺肌病模型小鼠的作用

摘　　要：目的探讨不同给药途径裘氏内异方治疗子宫腺肌病小鼠模型的作用。方法30只ICR雌性小鼠随机分成空白对照组、模型组、裘氏内异方口服汤剂组(汤剂组)、裘氏内异方颗粒制剂组(颗粒组)及裘氏内异方穴位贴敷贴组(贴敷组),每组各5只。HE染色观察小鼠子宫形态,免疫组化检测子宫中Ki-67、PCNA、MMP2和TIMP-2的表达水平,药物透皮示踪检测裘氏内异方透皮贴敷制剂透皮情况。结果模型组小鼠子宫可见大的病变结节和子宫肿胀,空白组与模型组小鼠子宫Ki-67、PCNA、MMP2、TIMP-2表达水平比较,差异有统计学意义(P<0.01);汤剂组、颗粒组和贴敷组小鼠子宫Ki-67、MMP2、TIMP-2表达水平低于模型组(P<0.05)。结论不同给药途径裘氏内异方均可以到达皮下组织发挥作用,其作用机制与降低Ki-67、PCNA、MMP2表达水平,升高TIMP-2表达水平有关。Objective To investigate the effects of different administration routes of Qiushi Neiyi decoction on the mouse model of adenomyosis.Methods Thirty female ICR mice were randomly divided into the blank control group,the model group,the Qiushi Neiyi decoction oral group(decoction group),the Qiushi Neiyi decoction granule preparation group(granule group)and the Qiushi Neiyi decoction acupoint application group(application group),with 5 mice in each group.The morphology of uterus was observed by HE staining,and the expression levels of Ki-67,PCNA,MMP2 and TIMP-2 in uterus were detected by immunohistochemistry.The transdermal effect of Qiushi Neiyi decoction was detected by drug transdermal tracing.Results Large pathological nodules and swelling were observed in the uterus of the model group.The expression levels of Ki-67,PCNA,MMP2 and TIMP-2 in the blank group were significantly different from those in the model group(P<0.01).The expression levels of Ki-67,MMP2 and TIMP-2 in the decoction group,granule group and application group were lower than those in the model group(P<0.05).Conclusion Qiushi Neiyi decoction can reach the subcutaneous tissue and play a role.The mechanism of action is related to the decrease of Ki-67,PCNA and MMP2 expression and the increase of TIMP-2 expression.
