摘 要:目的观察血液净化对重度中暑小型猪肠道组织闭锁小带蛋白-1(zonula occludens-1,ZO-1)和闭合蛋白(Occludin)表达的影响。方法构建小型猪重度中暑(heatstroke,HS)模型,血液净化干预治疗前后进行肠道组织功能评价。按照随机数字表法分3组:对照组(空白对照)、模型组(重度中暑模型)、干预组(血液净化治疗),每组各6只。HE染色观察肠道组织的病理形态变化;Chiu’s评分法评估肠道组织的损伤程度;免疫组织化学法(immunohistochemistry,IHC)观察肠道组织ZO-1和Occludin的表达。结果相对于模型组,干预组小型猪肠道组织细胞充血、水肿程度减轻,肠道黏膜结构较为完整。对照组、模型组和干预组的肠道Chiu’s评分比较差异有统计学意义(F=44.245,P<0.001);对照组、模型组和干预组的肠道组织ZO-1(F=152.477,P<0.001)和Occludin(F=40.360,P<0.001)比较差异有统计学意义。结论血液净化对重度中暑小型猪肠道损伤具有保护作用,可能与上调ZO-1和Occludin的表达有关。Objective To observe the effect of blood purification on the expressions of ZO-1(zonula oc-cludens-1)and Occludin in intestinal tissue of small pigs with severe heatstroke.Methods A model of severe heatstroke in small pigs was constructed,and the intestinaql function was evaluated before and after blood pu-rification intervention.According to the random number table method,the animals were divided into three groups:control group(n=6),blank control treatment was performed;model group(n=6),a model of heat-stroke was established;intervention group(n=6),treated with blood purification.Pathological changes of in-testinal tissue were observed using H-E staining.Chiu's score was used to assess the damage of intestinal tis-sue.Immunohistochemistry was used to observe the expressions of ZO-1 and Occludin in intestinal tissue.Results Reduced congestion and edema and relatively complete mucosa structure were found in intervention group,as compared with those in model group.Chiu's score was statistically different among the 3 groups(F=44.245,P<0.001).The expression levels of ZO-1 and Occludin were also statistically different among the 3 groups(ZO-1:F=152.477,P<0.001;Occludin:F=40.360,P<0.001).Conclusion Blood purification can be protective against intestinal injury due to heatstroke in small pigs,probably relating to the upregulation of ZO-1 and Occludin expressions.
