摘  要:[目的]利用黄芩药渣制备微生物菌剂,探究添加淡紫拟青霉发酵黄芩药渣(处理组,jd)与未添加淡紫拟青霉的黄芩药渣(对照组,ckj)的代谢差异,揭示淡紫拟青霉与黄芩药渣的互作发酵机制,为后续在大田中的推广应用提供理论依据。[方法]以未接种淡紫拟青霉菌株的黄芩药渣为对照,采用代谢组学技术分析淡紫拟青霉发酵黄芩药渣后的差异代谢产物和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes, KEGG)富集通路。[结果]对照组和处理组样本间的正负离子主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)均有良好区分,ckj和jd的代谢产物具有差异性,鉴定出429个上调代谢物和246个下调代谢物,KEGG代谢通路富集到29条差异显著的代谢通路。差异代谢产物主要为黄酮类、萜类、有机酸、生物碱、氨基酸和糖类,涉及到黄酮和黄酮醇生物合成,类黄酮生物合成,甘氨酸、丝氨酸、苏氨酸和色氨酸代谢,赖氨酸降解,苯丙氨酸、酪氨酸、色氨酸生物合成,半乳糖代谢,乙醛酸和二羧酸代谢及托烷、哌啶和吡啶生物碱的生物合成等8条差异富集通路。[结论]黄芩药渣经过淡紫拟青霉发酵后,萜类、有机酸、生物碱、氨基酸和糖类代谢物显著提高(P<0.05),这些物质为土壤养分含量的改善和植株免疫力的提高提供一定的营养基础。【Objective】Using Scutellaria baicalensis residue to prepare microbial agents,the present paper aimed to explore the metabolic differences between microbial agents prepared by fermentation of Scutellaria baicalensis residue by Paecilomyces lilacinus(jd)and the control without added strains of Scutellaria baicalensis residue(ckj)to reveal the mechanism of its interactive fermentation between strains and Scutellaria baicalensis residue,and provide theoretical basis for subsequent promotion and application in the field.【Method】Taking Scutellaria baicalensis residue without inoculated strains as the reference material,the differential metabolites and KEGG pathways of the fermentation of Scutellaria baicalensis residue by P.lilacinus were analyzed using metabolomics techniques.【Result】The PCA and OPLS-DA map of the positive ion and negative ion samples could be well distinguished.Volcano map analysis showed that the metabolite changes were different between ckj and jd,and there were 429 upregulated metabolites and 246 downregulated metabolites.According to the Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment metabolic pathway,29 significant metabolic pathways were obtained between ckj and jd.The differential metabolites mainly included flavonoids,terpenes,organic acids,alkaloids,amino acids,and sugars,which were annotated into eight metabolic pathways such as flavonoid and flavonoid biosynthesis,flavonoid biosynthesis,glycine,serine threonine and tryptophan metabolism,lysine degradation,phenylalanine,tyrosine,and tryptophan biosynthesis,galactose metabolism,glyoxylate and dicarboxylic acid metabolism,the biosynthesis of tropane,piperidine,and pyridine alkaloids,and so on.【Conclusion】Terpenes,organic acids,alkaloids,amino acids,and carbohydrate metabolites are significantly increased after the fermentation of Scutellaria baicalensis residue by P.lilacinus(P<0.05).These substances provide a certain nutritional basis for improvement of soil nutrient content and enhancement of plant immunity.
