麻黄-甘草药对水煎液对屋尘螨诱导的小鼠哮喘模型的影响及机制探究

摘　　要：目的研究麻黄-甘草药对水煎液对屋尘螨(HDM)诱导的小鼠哮喘模型的影响及其作用机制。方法将72只SPF级C57BL/6J雌性小鼠随机分成6组:正常对照组(Normal)、哮喘模型组(Model)、麻黄-甘草药对水煎液低剂量组(MG-L)、麻黄-甘草药对水煎液中剂量组(MG-M)、麻黄-甘草药对水煎液高剂量组(MG-H)和阳性对照地塞米松组(DXM),每组12只。采用HDM诱导建立小鼠哮喘模型,给予相应药物进行干预。分别运用瑞氏-吉姆萨染色分类计数BALF中细胞,肺组织H&E染色,ELISA法测定血清中IgE的含量和qPCR法检测肺组织中IL-4、IL-5、IL-13水平。结果麻黄-甘草药对水煎液高剂量组(MG-H)和阳性对照地塞米松组均可降低嗜酸性粒细胞的比例(P<0.01),同时提高肺泡巨噬细胞的数量(P<0.05);有效缓解HDM引起的肺部炎症症状,并在一定程度上恢复组织结构;有效抑制HDM刺激的IL-4、IL-5、IL-13的mRNA表达(P<0.05);降低血清IgE的分泌(P<0.005)。结论麻黄-甘草药对水煎液高剂量对屋尘螨诱导的小鼠哮喘模型具有明显改善作用,其作用机制与改善肺部异常免疫应答,抑制Th2型细胞因子和IgE的分泌,缓解肺部炎症和气道炎症反应等有关。Objective To investigate the effect of Mahuang-Gancao couple aqueous extract on mouse asthma model induced by house dust mite(HDM),and its mechanism of action.Methods 72 SPF grade C57BL/6J female mice were randomly divided into 6 groups:normal control group(Normal),asthma model group(Model),low-dose Mahuang-Gancao decoction group(MG-L),medium dose Mahuang-Gancao decoction group(MG-M),high-dose Mahuang-Gancao decoction group(MG-H),and positive control dexamethasone group(DXM),with 12 mice in each group.Establish mouse asthma model induced by House Dust Mite(HDM)and intervene with corresponding drugs.The cells in BALF were classified and counted by Wright-Giemsa Staining,H&E staining in lung tissue,the levels of IgE in serum were measured by ELISA,and the levels of IL-4,IL-5,IL-13 in lung tissue were detected by qPCR.Results MG-H and DXM all can significantly reduce the proportion of eosinophils(P<0.01),simultaneously increasing the number of alveolar macrophages(P<0.05);The lung inflammation caused by HDM was relieved effectively,and restored the organizational structure;The mRNA expression of IL-4,IL-5,and IL-13 stimulated by HDM were inhibited(P<0.05);The level of serum IgE was decreased(P<0.005).Conclusion Mahuang-Gancao couple aqueous extract can obviously improve the asthma model induced by house dust mite in mice by high dose of decoction,its mechanism is related to the improving abnormal immune response in the lungs,inhibiting the secretion of Th2 cytokines and IgE,alleviating lung inflammation and airway inflammatory response.
