摘  要:目的建立以对照药材为对照物质的定性和不依赖多种对照品定量的葛根药材“质-量”双标控制方法。方法“质”为采用高效液相色谱(HPLC)技术,以对照药材为对照物质建立特征图谱,以特征峰相似度明确药材真伪;“量”为对基准物质“大豆苷”定量,算出供试品中各特征峰相对含量,取“平均数-标准差”作为相对含量下限,依据特征峰相对含量下限明确葛根药材优劣。结果基于双波长等基线差示融合法建立的特征图谱和含量测定方法符合方法学考察要求;葛根对照药材与各供试药材的相似度均大于0.950;规定了葛根药材特征峰化学成分相对含量下限。结论该方法不依赖多种对照品,清晰、快速地判断药材的真伪优劣,为葛根药材的质量控制提供参考。Objective A qualitative and quantitative"quality-quantity"dual-standard control method for Puerariae Lobatae Radix medicinal materials based on the control materials as the control substance and the quantitative control method of Puerariae Lobatae Radix without relying on a variety of reference substances was established.Methods"Quality"refers to the use of high-performance liquid chromatography(HPLC)technology,using the control medicinal materials as the control substance to establish the characteristic map,and clarifying the authenticity of the medicinal materials by the similarity of characteristic peaks.The"amount"was the quantification of the reference substance"daidzein",the relative content of each characteristic peak in the test sample was calculated,and the"mean-standard deviation"was taken as the lower limit of relative content,and the advantages and disadvantages of Puerariae Lobatae Radix were clarified according to the lower limit of the relative content of characteristic peaks.Results The characteristic map and content determination method based on the dual-wavelength equal baseline difference fusion method met the requirements of the methodology.The similarity between the control and the tested herbs was greater than 0.950,and the lower limit of the relative content of characteristic peaks of the Puerariae Lobatae Radix was specified.Conclusion This method does not rely on a variety of reference substances,and can clearly and quickly judge the authenticity of medicinal materials,which provides a reference for the quality control method of Puerariae Lobatae Radix.
