摘  要:目的观察实验药物对大鼠脑组织水通道蛋白-4(AQP-4)mRNA表达的影响,探讨芳香开窍药对缺血性中风的作用机理,为临床上运用芳香开窍法提供实验依据。方法按Pulsinelli的4-VO法改良后建立大鼠全脑缺血再灌注模型,采用半定量RT-PCR方法检测大鼠脑组织中AQP-4mRNA表达。结果芳香开窍中药能降低缺血再灌注大鼠脑组织含水量;模型组AQP-4mRNA表达显著增加,其次是尼莫通组,芳香开窍中药组轻微减少。结论AQP-4可能参与缺血性脑血管病脑水肿的发生发展过程,AQP-4抑制剂可减轻脑水肿。尼莫通对AQP-4mRNA表达影响不大,芳香开窍药可降低AQP-4表达,从而减少脑水肿,这可能是其芳香开窍的机理之一。Objestive :To observe the effect of different dr ugs on the aquaporin -4of brain tissus and explore the therapeutic mechanism of inducing resuscitation method to ische mic stroke and provid e the experimental evidence for the clinic application thi s method.Methods:By estabilshing rat model of pulsinelli' s 4vo ischemia and reperfusion and ch ecking the expression of aquaporin -4mRNA in the rat' s bra in tissue by RT -PCR.Result:Drugs for inducing resuscitation could re duce th e water quantity in the brain t is-sue.There was a significant increase of the expression of aquaporin -4mRNA in the model group.Also there w as an increas e in Nimodipine group and slightly increase in herb g roup.Conclusion:Aquapori n -4may be involved in the appearance and dev elopmemt of ischemic brain edema ;aquaporin -4inhibitors can reduce t he brain edema.There is no great ef-fe ct on the expression of aquaporin -4mRna with Nmodipine;drugs for in-ducing resuscitation can decrease t he expression of aquaporin -4mRNA re-sulting in reducing the brain edema,which is possibly one of the mechanis m of inducing re suscitation.
