摘　　要：目的 :体外探讨ATRA和bFGF对成人骨髓间充质干细胞分化为神经元样细胞作用。方法 :采用Ficoll paque(1 0 77g/ml)分离液从骨髓分离出人骨髓间充质干细胞 ,体外扩增 ,以bFGF/ATRA为诱导剂 ,再加入BME、BHA、DMSO ,诱导期间观察细胞形态、数目的变化 ,并通过免疫细胞化学鉴定神经元烯醇化酶、神经丝蛋白、胶质纤维酸性蛋白的表达情况。结果 :免疫细胞化学显示在此诱导条件下 80 %的细胞表达NSE ,86 %的细胞表达NF M。结论 :ATRA和bFGF能诱导MSCs分化为神经元样细胞 。Objective:To investigate effect of ATRA and bFGF on adult mesenchymal stem cell differentiation into neuron like cell Methods:MSCs were isolated from adult bone marrow through density gradient with Ficoll paque (1 077 g/ml) and expanded in vitro MSCs were treated by ATRA and bFGF, then treated by BME, BHA, DMSOA Observe change of morphology and number of cell during differentiation Immunocytochemistry was used to detect the expression of NSE, NF M, GFAP Result:Under this condition, almost 80% of the cells expressed for NF M, 86% cell for NSE Conclusion: ATRA and bFGF can induce MSCs to differentiate into neuron like cell and can get higher percentage of induced differentiation than that of other reports
