俄罗斯大果沙棘组织培养技术研究

摘　　要：　主要以俄罗斯大果沙棘良种"优胜"的休眠枝茎段、幼嫩茎段为外植体,诱导形成不定芽及愈伤组织,同时对其他大果沙棘也进行了相关组织培养试验。结果表明,在诱导不定芽时,以休眠枝茎段为外植体,不定芽发生数量多,易获得无性系植株;基本培养基以1/2MS较好,继代培养以1/3MS为佳;最佳接种时间为3月份;初接种培养基为1/2MS+6-BA1.0mg/L+IAA0.5mg/L;愈伤组织诱导分化培养基为1/3MS+KT0.3mg/L+NAA0.03mg/L;增殖培养基为1/3MS+6-BA0.5mg/L+NAA0.2mg/L;生根培养基为1/4MS+NAA0.03mg/L+IBA0.1mg/L。Cluster adventitious buds and calluses were induced from dormant and growing stem segments of fine variety named 'YOUSHENG' of Hippophae rhamnoides L. from Russia.The tissue culture about other fine varieties can also be studied.The results showed that the amount of adventitious buds which were induced from dormant stem segments were more,the clone regenerated plants also were obtained easily.The basic culture medium is 1/2 MS medium,the basic multiplication medium is 1/3 MS medium.March is the best inoculation time.The inoculation medium is 1/2 MS+6BA 1.0 mg/L+IAA 0.5 mg/L,the callus inducing medium is 1/3 MS+KT 0.3 mg/L+NAA 0.03 mg/L,the multiplication medium is 1/3 MS+6BA 0.5 mg/L+NAA 0.2 mg/L,the medium for rooting is 1/4 MS+NAA \{0.03 mg/L\}+IBA 0.1 mg/L.
