中药消痹灵对软骨细胞一氧化氮合成的影响  被引量:9

摘  要:为探讨 NO与骨性关节突的关系及消痹灵 (XBL)治疗骨性关节炎的机理。取大耳白兔的软骨 ,用胶原酶分段消化法分离软骨细胞 ,分以下 4组培养。 1空白对照 :15 %血清的 DMEM。 2实验对照 :15 %血清培养液 (含 IL - 110 0 m g· ml- 1 )。 3消痹灵 组 :15 %血清培养液 (含 IL- 110 0 m g·m l- 1 5 % XBL)。4消痹灵 组 :15 %血清培养液 (含 IL- 10 0 mg· ml- 1 10 % XBL)。培养4 8小时后 ,测各孔亚硝酸盐浓度 ,换算出 NO浓度 ,结果显示 ,消痹灵能明显地抑制 IL- 1的作用 ,10 % XBL 优于 5 % XBL。表明消痹灵是通过抑制 IL~ 1的作用 ,降低了软骨细胞 NO的合成 ,从而减轻关节软骨的破坏 ,促进修复。Objective: In order to study the relation of NO and osteoarthritis(OA) and the mechanism of treating OA by Xiaobiling(XBL). Methods: Chondrocytes were isolated from Japanese white rabbits, and the method of 0.2% collagenase Ⅱ sequential digestion was used, four groups were divided as follows for culture: ① Control group, 15% serum DMEM. ②Model group, 15% serum DMEM+100ng/ml IL-1. ③XBL-Ⅰgroup, 15% serum DMEM+100ng/ml IL 1+5%XBL. ④XBL-groupⅡ, 15% serum DMEM+100ng/ml IL 1+10%XBL. Results: XBL can obviously depress the effect of IL 1 on chondrocytes, the effect of 10% XBL was higher than that of 5% XBL. Conclusion: XBL can decrease NO synthesis by depressing IL 1, therefore the destruction of articular chondrocyte was lessened and the repairation was promoted.
