摘 要:目的 研究低剂量钷 - 14 7(14 7Pm)内照射能否抵抗相继高剂量60 Coγ射线引起的细胞遗传损伤效应。方法 应用外周血红细胞 (RBC)计数 ,网织红细胞 (RC)计数 ,正常染红细胞 (NCE)微核和嗜多染红细胞 (PCE)微核检测 ,观察了低剂量14 7Pm内照射诱发小鼠红细胞微核的辐射交叉抗性。结果 剂量为 2 0Gy的60 Coγ射线外照可引起明显的外周血红细胞系统损伤 ,与正常对照组比较 ,RBC和RC计数显著减少(P <0 0 1) ;NCE和PCE微核细胞率明显增高 (P <0 0 1)。然而 ,预先应用低剂量14 7Pm 0 37,3 7,37Bq/g内照射预处理动物 ,3d后相继给予高剂量60 Coγ射线 2 0Gy外照射 ,RBC和RC计数显著增加 ,NCE和PCE微核细胞率显著降低 ,与单纯高剂量60 Coγ射线外照射相比较 ,差异有显著性或非常显著性 (P <0 0 5或P<0 0 1) ;其中某些指标与正常对照组比较 ,差异无显著性 (P >0 0 5 )。表明预先应用低剂量14 7Pmβ射线内照射动物 ,可使不同成熟期外周血红细胞对相继高剂量60 Coγ射线外照射引起的微核突变有了明显的抵抗力。低剂量14 7Pm诱导外周血红细胞系统的辐射抗性具有一定的剂量范围。结论 简便快速并适于进行连续观察的外周血红细胞微核可作为低剂量辐射生物效应的观察指标之一。Objective To explore whether low dose internal radiation of 147 Pm could act against subsequent high dose 60 Co γ radiation induced genetic damage in mice erythrocytes.Methods Counts of red blood cell(RBC),counts of reticulocytes(RC),micronucleated normochromatic erythrocytes(mn NCE) and micronucleated polychromatic erythrocytes(mn PCE)were measured and assayed in peripheral blood to observe the radiation crossing resistance of micronucleus mutation in mice erythrocyte induced by low dose 147 Pm internal radiation.Results A dose of 2 0 Gy 60 Co γ external radiation could lead to significant erythrocyte system injury.Compared with the control group,counts of RBC and RC decreased significantly(P<0 01)with significant increase of mn NCE and mn PCE(P<0 01).However,when treated with 0 37,3 7 and 37 Bq/g body weight of 147 Pm internal radiation beforehand,and 3 days later followed by 2 0 Gy 60 Co γ external radiation,compared with 60 Co external γ ray radiation only,counts of RBC and RC increased significantly with significant decrease of mn NCE and mn PCE(P<0 05,or P<0 01).Some indices even showed no significant difference compared with the control group with any radiation at all(P>0 05).It suggested that prior treatment with 147 Pm internal radiation could improve the resistance of micronucleus mutation in erythrocytes at different mature stage induced by subsequent 60 Co γ external radiation.There existed a dose range for inducing the radiation crossing resistance by low dose 147 Pm internal radiation.Conclusions Genetic mutation in peripheral blood erythrocytes with characteristics of simple,quick and suitable for continuous observation might be one of the biological indexes for low dose radiation effect.
