摘 要:目的:探讨维A酸类药物对人类皮肤光老化的拮抗作用及其可能作用机制。方法:以PUVA治疗的非暴露部位皮肤建立人类皮肤光老化模型,采用HE染色、Verhoeff染色、电镜、酶组织化学、免疫组织化学等方法检测PUVA治疗及口服芳维A酸乙酯对多项皮肤光老化相关指标的影响。结果:PUVA治疗后,银屑病背部非皮损区皮肤组织学和生物学特性上迅速出现具有光老化特征性的改变,真皮成纤维细胞由分裂表型转化为无分裂活性表型,B1、B2、C1、C2各组衰老相关β-半乳糖苷酶和P16蛋白表达阳性率分别为42.9%,100%,15.4%,23.1%和71.4%,100%,30.8%,53.8%;各组比较均有显著性差异(P<0.001)。结论:口服维A酸类药物对PUVA诱导的皮肤光老化有明显的拮抗作用,其作用机制可能与其调控真皮成纤维细胞衰老相关的基因表达有关。Objective:To investigate the antagonism of ora l retinoids to skin photoaging induced by photochemotherapy with psoralnes and ultraviolet A(PUVA)and its possible mechanism.Methods:The model of skin photoaging of human was established by non -lesion back skin of vulgar psoriatic patien ts with PUVA treatment ,HE stain,Verhoeff stain,electron microscope,e nzyme cytochemistry and immunocytochemistry were used to study influences of PUVA and oral retin oids on the skin photoaging -characteristic biological markers.Results:After treatment with PUVA,the sampl es of B group displayed histological and biologic characteristics of ski n photoaging rapidly,the positive rates of senescence -associated -β-galactosidase and P 16 protein of B,C1,C2group were 42.9%,100%,15.4%,23.1%and 71.4%,100%,30.8%,53.8%respec tively,there were significant differences among each group(P<0.001).Conclusion:Oral retinoids had prominent antagonism to skin photoaging,its mechanism ma y due to the regulation of senescence-associated genes of dermic fibroblasts
