活血、益气方药对心梗后左心衰大鼠血TNF和血小板聚集影响的对比研究

摘　　要：目的:对比观察活血、益气方药对心肌梗死后左心衰大鼠血肿瘤坏死因子(TNF)和血小板聚集的影响。方法:采用大鼠心梗后左心衰动物模型,比较观察由丹参、川芎、红花组成的活血注射液和由党参、黄芪组成的益气注射液对心梗后左心衰大鼠血TNF、血小板聚集等指标的影响。结果:模型组较假手术组血TNF水平明显升高(2.842±0.983;1.412±0.387,P<0.01),各治疗组均较模型组明显下降(P<0.05);模型组与假手术组比较,血小板聚集率显著升高(67.06±3.95;46.33±3.56,P<0.05),各治疗组治疗后血小板聚集率较模型组降低(P<0.05或P<0.01)。结论:活血、益气方药通过减少TNF的分泌或拮抗TNF的途径而降低血浆TNF水平,这可能是益气、活血方药改善心衰血流动力学、干预心室重构、抑制心肌细胞凋亡的生化机制之一。Objective:To observe the effects of the herb of activating blood,nourishing qi on tumor necrosis factor(TNF) and platelet aggregation of rats with left heart failure after myocardial infarction. Methods:The rat model with left heart failure after myocardial infarction was adopted. These rats were treated with activating blood injection consisting of Chuanxiong rhizome and Red Rooted Salvia and Nourishing Qi injection consisting of Salvia miltiorrhiza and milkvetch. TNF and platelet aggregation were detected. Results:TNF in the model group (2.842±0.983) was higher significantly than the control group (1.412±0.387,P<0.01), but TNF in every treated groups were lower significantly than the model group(P<0.05);Compared with the control group, the platelet aggregation rate of the model group was significantly raised (67.06±3.95;46.33±3.56, P<0.05), while that of every treated groups were lower significantly than the model group(P<0.05 or P<0.01). Conclusion:The herb of activating blood,nourishing qi could lower TNF in plasma through decreasing TNF secretion or prohibiting TNF. It may be one of biochemical mechanism of activating blood and nourishing qi herb improving hemodynamic,preventing and reversing left ventricular remodeling,inhibiting myocardial cell apoptosis. Both the herb of activating blood and the herb of nourishing qi could improve the platelet aggregation of rats with heart failure. This reflect TCM correlation theory of Qi and blood. It may be one of mechanism of the herb treating heart failure.
