摘 要:目的 探讨He Ne激光照射大鼠心前区对心肌毛细血管通透性及毛细血管内皮细胞血管内皮生长因子 (VEGF)表达的影响。方法 Wistar大鼠随机分为对照组和激光照射组 ,每组各 8只 ,其中照射组He Ne激光照射剂量为 60 5J cm2 ,垂直照射大鼠心前区 ,每日 1次 ,共 1 0次。采用铁蛋白大鼠静脉灌注示踪 ,心肌标本于透射电镜下观察和免疫组织化学染色图像分析的方法 ,观察心肌毛细血管通透性及VEGF变化。结果 电镜下 ,激光照射组心肌内层毛细血管管壁内、外以及心肌细胞的线粒体内均可见大量的高密度铁蛋白颗粒 ,较对照组明显增多 ;免疫组化染色示照射组心肌内层毛细血管内皮细胞的着色明显加重 ,其平均光密度值为 0 2 63± 0 0 1 5 ,明显高于对照组的 0 2 2 7± 0 0 1 6 (P <0 .0 1 )。结论 以剂量为 60 5J cm2 的He Ne激光照射大鼠心前区 ,可使心肌毛细血管的通透性和VEGF的表达明显增强 ,为He Ne激光照射能改善心肌微循环提供了实验形态学依据。Objective To study the change in capillary permeability in myocardium and the expressions of VEGF in endothelium of myocardial capillaries by He Ne laser irradiation on precoadialc region of Wistar rat. Methods Sixteen rats were equally divided into control and irradiated groups. The dose of 60 5 J/cm 2 was used every day in irradiated group. After 10 days, the ferritin was perfused into femoral vein and the capillary permeability was observed using electron microscope. The immunohistochemical method and computer image analysis were applied in the experiment. Results The ultrastructure of the myocardial capillary was studied. A lot of high density granules in the capillary endothelium, out of capillary and in the mitochondria of myocardial cells were seen in the internal layer of myocardium of irradiated group. In control group, granules were a few. The staining color by immunohistochemistry of VEGF in the internal layer of myocardium in irradiated group were heavier than in control one. The mean optic density values in irradiated group(0 263±0 015) were higher than that of control one(0 227±0 016, P<0 01) in the internal layer of myocardium. Conclusion The results indicated that myocardial capillary permeability and the expression of VEGF in the internal layer of myocardium endothelium were increased after He Ne laser irradiation. The study provided experimental morphological evidences that myocardial microcirculation could be improved by He Ne laser irradiation.
