摘  要:目的 研究NMDA受体亚单位 (NR1、NR2A和NR2B)mRNA在正常成年大鼠海马结构各区的表达及形态特点。方法 原位核酸分子杂交组织化学反应和图像处理与分析。结果 NR1、NR2A和NR2BmRNA在正常大鼠海马结构各区锥体细胞和颗粒细胞普遍表达 ,其中NR1mRNA的表达最强 ,NR2AmRNA的表达最弱。NR1mRNA在齿状回 (DG)的表达水平明显强于CA1区和CA3区 ;NR2AmRNA在CA1区的表达水平则强于CA3区和DG ;NR2BmRNA在海马各区的表达水平基本相同。结论 NR1、NR2A和NR2BmRNA在正常成年大鼠海马各区的表达模式不同 ,其中NR2AmRNA在海马CA1区以及NR1mRNA在DG高表达的分布特点可能与海马的学习。Objective To investigate the mRNA expression of NMDA receptor subunits (NR1, NR2A and NR2B) in adult rat hippocampus.?Methods In situ hybridization histochemistry staining and LEICA QWin image processing and analysis system were used in the study.?Results The mRNA of NMDA receptor subunits NR1, NR2A and NR2B was widely distributed in the pyramidal and granular cells in all hippocampal subfields. The expression of NR1 mRNA was the strongest, and that of NR2A mRNA was the weakest. The expression of NR2B mRNA was about the same in the different hippocampal subfields, while that of NR1 mRNA was stronger in the dentate gyrus than in the CA1 and CA3 subfields, and that of NR2A mRNA was stronger in CA1 than in CA3 and dentate gyrus.?Conclusion The mRNA of NMDA receptor subunits NR1, NR2A and NR2B are expressed in different patterns in normal adult rat hippocampus. The higher expression of NR2A mRNA in CA1 subfield and that of NR1 mRNA in dentate gyrus may be related to the physiology of learning and memory and to the selective vulnerability to ischemic injury of the hippocampus.
