摘  要:目的 探讨泼尼松龙鞘内注入镇痛的脊髓机制。方法 慢性鞘内置管的SD大鼠随机分为泼尼松龙组和对照组 ,每组 6只。泼尼松龙组大鼠鞘内注入泼尼松龙 2mg kg(2 0 μl) ,对照组注入等容积生理盐水 ,1h后进行福尔马林试验 ,动物继续存活 2h后取脊髓腰段 (腰膨大 )多聚甲醛固定的冰冻切片。P物质免疫组化显示用ABC法。结果 两组大鼠右侧脊髓背角浅层P物质光密度值较左侧明显增加 ,泼尼松龙组大鼠脊髓背角浅层P物质含量较对照组无明显差异 (P>0 .0 5 ) ,P物质免疫反应产物无明显变化。Objective To study the effects of intrathecal prednisolone acetate on the substance P (SP) in spinal cord of rat and the spinal mechanism of its pain relief. Methods Chronic lumbar intrathecal catheters were implanted in SD rats and these rats were randomly divided into two groups: prednisolone group and control group (n=6 for each). Fomalin test in the right hind sole was carried out 1 h after the intrathecal administration of prednisolone (2 mg/kg) or nomal saline. The animals were sacrificed 2 h later to measure SP in the slices of rat spinal cord by using immunohistochemical ABC technique. Results The optical density (OD) of immunoreaction of SP in the superficial layers of the right spinal dorsal horn was significantly higher than that of the left spinal dorsal horn. No difference in the content of SP in the superficial layers of spinal dorsal horn was noticed between the 2 groups (P>0.05). Conclusion Intrathecal prednisolone acetate would have no effect on the production of SP in spinal cord in response to nociceptive stimulation.
