益气化瘀方对腰神经根压迫模型神经肌肉接合部施旺细胞的作用

摘　　要：目的 :研究大鼠 L5神经根受压后 ,益气化瘀方对神经肌肉接合部施旺细胞的作用和表现。方法 :采用免疫组织化学和激光共聚焦扫描技术 ,使用 S-1 0 0多克隆抗体标记施旺细胞 ,α-bungarotoxin (α-BTX)萤光结合剂显示运动终板。大鼠 L5神经根受压后 ,给予益气化瘀方治疗 ,于术后 1 0、2 0和 3 0天取比目鱼肌 ,观察神经肌肉接合部施旺细胞在神经再生过程中的表现。结果 :肌肉失神经支配后 ,益气化瘀组的末梢施旺细胞在神经肌肉接合部的聚集 ,出芽数量及突起的延伸长度均明显优于对照组 ,神经再生期间 ,末梢施旺细胞与运动终板的接触面及肌肉接合部的再构筑 ,益气化瘀组亦同样明显优于对照。结论 :益气化瘀方能促进施旺细胞的增生及提高其再生功能 ,加快神经肌肉接合部的重建 ,缩短神经再生修复进程。Objective:To study the effect of the YI QI HUA YU recipe on Schwann cells in a neuromuscular junction after compression of the L5 nerve root in rats.Methods:Using immunohistochemistry, confocal laser scanning techniques and the polyclonal antibody S-100 as a Schwann cell marker, motor endplates were visualized with fluorescein-conjugated a-bungarotoxin(a-BTX). The effects of the YI QI HUA YU recipe on Schwann cells in a neuromuscular junction were observed at 10d? 20d? 30d?and 60d after the L5 nerve root was compressed.Results:The aggregates ,sprouts and extension of terminal Schwann cells during degeneration, the amount of overlap of terminal Schwann cells with endplates and the reformation of neuromuscular junctions during regeneration were superior in cells treated with the YI QI HUA YU recipe than in control cells. Conclusion:The YI QI HUA YU recipe can promote the increase of Schwann cells and enhance regeneration. It can also accelerate the reformation of neuromuscular junctions and reduce regeneration time.
