摘  要:研究 β 淀粉样蛋白 (β AP)对衰老大鼠海马神经元的影响。采用D 半乳糖 (D gal)建立衰老动物模型 ,海马内微注射 β AP ,检测各组大鼠Y 迷宫分辨学习和一次性被动回避反应的变化 ,并测定海马超氧化物歧化酶 (SOD)活性、丙二醛 (MDA)含量、Na+ K+ ATP酶活性和海马线粒体膜流动性。可见 β AP加上D gal组大鼠学习记忆能力 ,与只用 β AP或D gal组比较有显著性差异 ;与 β AP或D gal组比较Na+ K+ ATP酶活性显著降低、MDA含量显著增加、海马线粒体膜流动性显著降低。结果表明 ,β AP与D gal联合使用可导致衰老大鼠脑海马自由基损伤加重 ,学习记忆能力显著减弱 ,To study the effect of β amyloid protein(β AP) on the neurons of hippocampus in the aging model of rats. Based upon the model of aging rats induced by D galactose(D gal) and hippocampal microinfusion of β AP, the behaviors of rats were measured by Y maze and one trial passive avidance response, SOD and Na + K + ATPase activity. The content of MDA and the viscous coefficient(η)of mitochondrial membrane were measured. In the D+A rats, the abilities of learning memory were remarkably attenuated as compared with other groups( P <0.05 , P <0.01 ); SOD and Na + K + ATPase activity was markedly lower while the content of MDA was increased markedly. The fluidity of mitochondrial membrane was remarkably lower than N group( P <0.01). The result suggested that the combination of D gal and β AP could induce the increase of damage by free radical in rat hippocampus and a conspicuous reduction of learning memory ability in rat.
