摘 要:目的探讨慢性缺氧膈肌被动收缩特性与细胞骨架结构及基因表达的关系。方法用慢性间断缺氧方法制备慢性缺氧动物模型,测定膈肌条的应力-应变关系,免疫组化和RT-PCR方法检测膈肌细胞骨架蛋白titin,nebulin的表达。结果(1)慢性缺氧组膈肌肌肉僵硬度常数下降,初始弹性常数升高,与对照组相比差异有显著性意义(P<0.05);(2)慢性缺氧膈肌细胞titin,nebulin表达显著减少,与对照组相比差异有显著性意义(P<0.05);(3)相关分析显示,膈肌细胞骨架蛋白表达的变化与膈肌肌肉僵硬度常数变化相关。结论慢性缺氧大鼠膈肌细胞titin和nebulin表达下调,并可能与膈肌被动张力改变有关。Objective To invstigated the relationship between the expression level of cytoskeleton and passive tension of diaphragm.Method Experimental models were induced by discontinuous hypoxia and the stress strain relationship of muscle strips were detected.Immunocytochemistry and RT PCR procedures were used to analyze the expression of two key elastic components of muscle , titin and nebulin.Result (1)Muscle stiffness constant was lower and muscle elastic modulus was higher in hypoxia rats than that in controls(P< 0.05).(2) Compared with controls,exprssion of titin and nebulin in diaphragmatic cells of hypoxia rats was lower(P< 0.05).(3) The dexrease of expression of titin and nebulin was consistent with the decline of the stiffness constant of muscle.Conclusion Expression level of titin and nebulin in diaphragmatic cell of hypoxia rats decreases significantly.And it may associate with the decline of passive tension of diaphragm.
