摘 要:目的利用正交设计优化当归多糖水提取工艺。方法采用L9(33)正交设计方法,取3个因素即水提取过程中的加水量(A)、提取时间(B)、提取次数(C),每个因素3个水平依次试验,醇沉工艺按照文献将水提液浓缩至1∶1.8,加95%乙醇至醇浓度为65%。最终采用苯酚-硫酸法测定当归多糖的含量,以当归多糖的得量作为考核指标对9次试验结果进行评价,考核本试验的可靠性并进行验证。结果影响当归多糖得率最主要的因素为提取时间。当归多糖得率最优的提取工艺为10倍量水,提取时间为60 min,提取3次;相关数据显示本次研究的精密度、稳定性均较好,验证结果基本与正交试验设计分析结果一致。结论当归多糖得率最优的提取工艺为加10倍量水、提取60 min、连续提取3次。Objective To optimize the extraction technology for Angelica sinensis polysaccharide( AP) under the orthogonal design. Methods Three factors: added water( A),extraction duration( B) and extraction time( C) in three levels were tested orderly. Alcohol deposition method was used to concentrate water extract to 1 ∶ 1.8.95% alcohol was added to get concentration of 65%. The concentration of AP was determined by phenol-sulfuric acid method. AP quantities received in 9 tests were evaluated as well as the reliability of this experiment was assessed to inspect and verify. Results B was the main factor in influencing the yield of AP. The best technology was A3B1C3. The data showed that the maximum yield was got in 8thtest basically in accord with the analysis. The experiment was precise and stable. Conclusion The best extraction technology for AP is following: 10-fold added water,60 minutes extraction duration and 3 extraction times in a row.
