摘 要:红细胞生成素(EPO)在临床和基础研究中应用广泛。本文用从人尿中提取的EPO免疫动物制备出两株分泌抗EPO单克隆抗体的杂交瘤细胞系,分别命名为HIEPO2和HIEPO11。经鉴定HIEPO2和HIEPO11均能与重组人EPO(rHuEPO)反应,其免疫球蛋白类型均为IgM,用HIEPO2和HIEPO11制备的亲和层析柱纯化人尿中EPO,前者效果优于后者。HIEPO2对EPO的活性有抑制作用,HIEPO11对EPO的活性无影响。Erythropoietin ( EPO ) is widely used in clinical and basic studies. In the present experiment, two hybridoma cell lines (HIEPO2 and HIEPO11 ) secreting monoclonal antibody against EPO were produced by fusion of mouse myeloma cells ( NS1 ) and spleen cells from BALB/c mice immunized with EPO purified from human urine. It was found that both the McAbs were bound to recombinant human EPO arid their immunoglobulin isotype subclass was IgM.. The quantity and toxicity of EPO purified by HIEPO2 were greater than that byHIEPO11, Moreover, HIEPO2 may suppress the activity of EPO while HIEPO11l cannot.
