摘 要:真核细胞中,各细胞器之间的物质交流主要是通过膜泡运输完成的。膜泡运输依赖多种跨膜蛋白和可溶性蛋白的协同作用来完成。我们前期研究发现的拟南芥MAG2是在高尔基体与内质网之间的膜泡运输过程中起作用的拴留因子,MAG2与MIP1、MIP2和MIP3形成拴留复合体,对种子储藏蛋白从内质网的运离起重要的调控作用。为了深入了解MAG2参与的膜泡运输途径的分子机制,我们制作了mag2-1xmip3-1双重突变体。通过一系列观察分析发现,当MAG2和MIP3同时缺失,不仅种子储藏蛋白质的运输受到严重阻碍,植物的生长发育也受到严重影响,对环境条件的变化更加敏感。研究结果表明,MAG2复合体不仅调控蛋白质的运输,还对植物的生长发育有重要调控作用。In eukaryotic cells, material transport between organelles is completed mainly by vesicle trafficking.Vesicle transport dependents on the synergistic function of a variety of transmembrane proteins and soluble proteins. Our previous study found that Arabidopsis MAG2 is a tethering factor functions between the ER-Golgi.MAG2, MIP1, MIP2 and MIP3 form a tethering complex that plays an important role on exit of seed storage proteins from the ER. To clarify molecular mechanism underlying MAG2-involved transport pathways, we made mag2-1xmip3-1 double mutant. Through a series of observation and analyses, we found that simultaneous d epletion of MAG2 and MIP3 not only block exit of the seed storage proteins transport, but also affect plant growth and development. The mutant plant is more sensitive to environmental stress. The results indicate that in addition to regulation of transport of proteins, MAG2 complex is also involved in the growth and development of plants.
