摘 要:为研究和挖掘贵州地方特色猪种贵州白香猪的遗传种质资源,本实验克隆了该猪种的白细胞介素2(IL-2)、IL-4、IL-6基因,并对其相关遗传信息进行了分析。克隆测序结果表明,IL-2、IL-4和IL-6的ORF大小分别465 bp、402 bp和639 bp,其与猫、牛、狗、马、人、猴、鼠、兔、羊的推导氨基酸的序列相似性分别为57.8%~74.7%、47.8%~87.6%和45.3%~76.4%,与已报道的其它猪种的推导氨基酸的序列相似性均在98%以上。不同猪种间,除IL-4推导氨基酸序列完全相同外,IL-2的差异主要出现在第2位,第87位以及第125位,IL-6则仅出现在第30位和第69位。推导蛋白质特性分析结果表明IL-2、IL-4和IL-6大小分别为17 401.2 Ku、15 022.6 Ku、23 880.5 Ku,均系亲水性蛋白质,信号肽长度分别为20、24、25个氨基酸残基,分别含有27、26和35个磷酸化位点,仅IL-6在7~26位氨基酸之间含有一个跨膜螺旋区。不同猪种间,尽管上述白细胞介素在推导氨基酸水平均存在一定差异,但这些差异并未引起其预测模型空间结构改变,表明其在功能上具有高度保守性。In order to research and excavate the Guizhou Baixiang pig genetic germplasm resources of characteristic pig breed of Guizhou, the IL-2, IL-4 and IL-6 genes of Guizhou Baixiang pign were cloned and analyzed. The sequencing results show that the ORF of IL-2, IL-4 and IL-6 is 465 bp, 402 bp and 639 bp,respectively. The sequence similarity of amino acid derived with cat, cattle, dog, horse, human, monkey, mouse,rabbit and sheep is 57.8%~74.7%, 47.8%~87.6% and 45.3%~76.4%, and with the other pig breeds is beyond 98%,respectively. Except the amino acid sequence of IL-4 is identical, the differences of IL-2 are at the 2 th, 87 th and125 th amino acid, and the IL-6 only at the 30 th and 69 th, among different pig breeds. The protein structure analysis results showed that the weight of IL-2, IL-4 and IL-6 is 17 401.2 Ku, 15 022.6 Ku, 23 880.5 Ku, which are Hydrophilic protein with 27, 26 and 35 phosphorylation sites. The length of the signal peptide is 20, 24 and25 amino acids, respectively, and the transmembrane helical segment only exist in the IL-6, which is between 7 th^26 th amino acid. Although, there are some differences in the amino acid of IL-2 and IL-6, they don't cause any distinctions in space structure predicted. The results suggest that all of them have high conservatism in the biological function.
