摘 要:目的:观察汉防己甲素(tet,tetrandrine)对低温灌洗保存大鼠供肝的肝细胞内Ca2+浓度及肝活性的影响.方法:选择SD大鼠并分组:A组术前腹腔内注射生理盐水;B组术前腹腔内注射Tet;C组术前腹腔内注射尼膜地平;各组均以4℃乳酸林格液进行供肝灌洗并低温保存2h.然后检测各组的高能磷酸化合物及肝细胞内Ca2+浓度,并同时观察了肝脏的病理改变.结果:应用Tet组,肝脏的能量代谢、肝细胞内Ca2+超载及病理变化均有明显改善.结论:Tet可减少低温灌洗保存大鼠供肝的肝细胞内Ca2+超载,降低肝细胞的能量损耗,从而增强供肝的活力.AIM:To study effects of tetrandrine (Tet) on the densityand activity of Ca2+ in the liver cell during cold storage.METHODS:SD rats were divided into three groups. Beforeoperation, Group A was injected saline, Group B, Tet , andGroup C, nimodipine into the abdominal cavity. The liversof all the animals were perfused with lactic acid Ringer’ssolution at 4 ℃ and then stored for 2 hours in hypothermia.High energy phosporic acid compound and the density ofCa2+ in the liver cells were detected in all the groups; Thepathologic changes were observed under microscope.RESULTS:The energy metabolism and density of Ca2+ inthe liver cells and pathologic changes in the group B weremuch well improved than those in the other groups.CONCLUSION:Tet can decrease the overloading of Ca2+ in theliver perfused in hypothermia, lower the energy loss of the livercell, and so strengthen the activity of transplanted liver.
