柔肝冲剂抗肝纤维化的动物实验

摘　　要：为研究柔肝冲剂(主药:黄芪、丹参、郁金)抗肝纤维化的作用,以Wistar雄性大鼠腹腔注射二甲基硝胺制作肝纤维化模型。观察柔肝冲剂对模型鼠纤维化相关指标的影响。结果:柔肝冲剂能抑制模型鼠肝脏胶原蛋白的合成;病理切片显示,柔肝冲剂有减轻肝脏炎症反应,保护肝细胞作用;NOrthem印迹分子杂交结果显示,柔肝冲剂能降低模型鼠纤维肝α_1(Ⅰ)、α_1(Ⅲ)和α_1(Ⅳ)前胶原mRNA的表达。提示:柔肝冲剂能同时影响肝脏间质细胞和实质细胞胶原基因的表达,减少胶原的生成,从而抑制肝纤维化的发展。In order to study the anti - hepatic fibrosis of 'Liver - Softening Granules' (main ingredients: Radix Astragali seu Hedysari, Radix Salviae Miltiorrhizae, Radix Curcumae), hepatic fibrosis model was made by injection of dimethyl nitroamine into the abdominal cavity of Wistar male rats, to observe influence of 'Liver- Softening Granules on the related indexes of fibrosis in the model rats. The results showed that 'Liver- Softening Granules' could inhibit synthesis of collagen in the h'ver of the model rats. The pathological sections showed that ' liver - Softening Granules' possesses the functions to subdue the inflammatory reaction of the liver and protect the hepatic cells. The results in Northern molecule hybridization showed that liver- Softening Granules can decrease the procolla-gen mRNA expression of fibrous liver (1(Ⅰ) , (1(Ⅲ) and (1(Ⅳ) in the model rats. The findings indicate that 'Liver -Softening Granules' can simultaneously influence the collagen gene expression of the interstitial cells and parenchymal cells of the liver and decrease the formation of collagen, so as to inhibit the progress of the hepatic fibrosis.
