白消安诱发造血功能障碍小鼠模型的建立及淋巴细胞转化功能的测定

摘　　要：目的　建立白消安诱发急性造血功能障碍的动物模型 ,探讨淋巴细胞在骨髓再生障碍中发生变化的基础和意义。　方法　采用白消安悬液 (2 m g/ml)每天两次胃饲小鼠 ,每次 0 .2～ 0 .3ml,胃饲 7～ 10天 ,观察其外周血和骨髓液中细胞数量和形态的变化 ,并检测外周血淋巴细胞转化功能。　结果　用该方法诱导小鼠后 ,其红细胞、白细胞、血小板、网织红细胞及骨髓有核细胞计数均明显减少 (P<0 .0 1) ,骨髓象检查显示骨髓正常造血组织被脂肪细胞等替代。外周血淋巴细胞转化率明显升高 (P<0 .0 1)。　结论　白消安诱导小鼠急性造血功能障碍结果稳定 ,发生率高 ,方法简单 ,各项指标与人的再生障碍性贫血相似 ;Objective\ To establish a simple animal\|model with hematopoietic stem cell failure and explore the significance of the changes of lymphocyte.\ Methods\ Each medicine\|induced mouse was \{gastric\} fed with myleran(0 2 mg/ml) twice a day(0 2 or 0 3 ml once) in 7~10 days until the symptom arose and the transform ratio of T lymphocyte in the mice blood was calculated.\ Results\ The amount of erythrocyte, leukocyte, thrombocyte, reticulocyte and the cell with nuclear in mice marrow was greatly lower than the mice in control group(P<0 01).\ The marrow examination showed that the bone marrow was replaced by fat cells.\ The transform ratio of lymphocyte in blood raised greatly(P<0 01).\ \{Conclusion\}\ The method of the mouse model with hematopoietic stem cell failure induced by myleran is simple.\ The symptom is stable and all the indices is exact.\ The abnormality of lymphocyte multiplication may relate to hematopoietic aplasia in mouse.\;
