摘  要:目的 探讨亚慢性砷中毒大鼠肝脏中环核苷酸变化及其与脂代谢紊乱的关系。方法 采用亚慢性毒性试验方法 ,大鼠经口灌胃染毒 ,试验期为 6周。实验结束后取肝脏测腺苷环一磷酸 (cAMP)、鸟苷环一磷酸 (cGMP)、一氧化氮 (NO)和丙二醛 (MDA)。结果 染砷大鼠肝脏中cAMP、cGMP含量均显著高于对照组 (P <0 .0 5 ) ,且肝脏中NO、MDA含量明显升高 ,有剂量反应关系 ,与对照组相比有显著差异 (P <0 .0 5 )。结论 亚慢性砷中毒大鼠第二信使系统平衡紊乱 ,肝脏内NO、MDA含量增多可能与脂质的分解、合成、转运等多环节出现障碍有关。Objective To observe the changes of cyclicnucleotide in subchronic arsenic-poisoning rat livers and explore its relations to abnormal lipid metabolism. Methods Subchronic toxicity test was applied. The rats were exposed to arsenic by oral perfusion at gradient doses for 6 weeks respectively. After the rats were sacrificed,cAMP,cGMP,NO and MDA were detected in the liver homogenate. Results The levels of cAMP and cGMP were significantly higher in the liver of arsenic-poisoning rats than in control group(P<0.05). And compared with that of control group,the contents of NO and MDA in the liver homogenate of arsenic-poisoning rats remarkably increased(P<0.05)and there was the dose-effect relationship. Conclusion The balanceof the second messenger is broken and the remarkable increases of contents of NOand MDA may play a crucial role in the abnormal lipid metabolism .
