摘　　要：目的采用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱法(UHPLC-Q-Exactive Focus-MS/MS)对陕产附子中的生物碱类成分进行分析和鉴定。方法采用Waters Acquity UPLC BEH C_(18)色谱柱(2.1 mm×50 mm,1.7μm),以乙腈-0.1%甲酸水溶液进行梯度洗脱。使用电喷雾离子源(ESI),正离子模式扫描,扫描范围m/z 100~1500。结果通过二级高分辨质谱分析结合对照品及相关文献,共鉴定出228个生物碱类成分,包括C_(18)类21种、C_(19)类145种[双酯型生物碱(DDAs)42种、单酯型生物碱(MDAs)47种、醇胺型生物碱(ADAs)42种、脂类(Lipo-As)14种]、C_(20)类27种以及其他35种,其中28个化合物为首次从附子或该植物中鉴定出。结论对附子生物碱类成分进行了较为全面系统的解析,为后续药效物质基础研究及应用开发奠定基础。Objective Ultra performance liquid chromatography coupled with quadrupole/electrostatic field orbital trap high resolution mass spectrometry(UHPLC-Q Exactive Focus MS/MS)method was developed to rapidly analyze and identify the alkaloids from Shaanxi-produced crude Fuzi(Aconiti Lateralis Radix Praeparata).Method The chromatographic separation was performed on the Waters Acquity UPLC BEH C_(18)column(2.1 mm×50 mm,1.7μm)with acetonitrile-0.1%formic acid water solution as the mobile phase for gradient elution.The electrospray ion source(ESI)was adopted to collect the data in positive ionization modes,and the detection range was m/z 100~1500.Results The chemical constituents were identified and confirmed by analyzing the mass data from the reference substances and literature.A total of 228 alkaloids were identified,including 21 C_(18)alkaloids,145 C_(19)alkaloids[42 diester diterpenoid alkaloids(DDAs),47 monoester diterpenoid alkaloids(MDAs),42 aminoalcohol-diterpenoid alkaloids(ADAs),14 lipo alkaloids(Lipo-As)],27 C_(20)alkaloids and 35 others,28 of them were discovered for the first time.Conclusion The established method systematically analyzed the alkaloids of Shaanxi-produced crude Fuzi(Aconiti Lateralis Radix Praeparata).It provides a reference for the subsequent study of material basis and development of Shaanxi-produced crude Fuzi(Aconiti Lateralis Radix Praeparata).
