摘 要:目的探究麦冬皂苷D(OP-D)对于小鼠压力超负荷病理性心肌肥厚的保护作用。方法运用主动脉弓缩窄术(TAC)建立小鼠压力超负荷病理性心肌肥厚。C57/BL6J小鼠随机数字法平均分为4组:假手术+溶剂组(Sham+Vehicle);假手术+麦冬皂苷D组(Sham+OP-D)、手术+溶剂组(TAC+Vehicle)、手术+麦冬皂苷D组(TAC+OP-D),每组8只。经由腹腔注射的方式给予OP-D。通过小动物超声检测左心室射血分数(LVEF)和短轴缩短率(FS)评估小鼠心脏功能;通过心脏舒张末室间隔厚度(IVSd)和舒张末左心室后壁厚度(LPWd),心脏重量,心脏重量与胫骨长度比值(HW/TL),小麦胚芽凝集素染色计算心肌细胞平均横截面积(CSA)评估小鼠心脏病理性肥厚;通过马松染色检测小鼠心脏纤维化水平;最后运用定量聚合酶链反应对肥厚相关分子Nppa、Nppb、Myh7和纤维化相关分子Tgfb1、Col1a1和Col1a3的转录水平进行检测。结果经过OP-D治疗后,与TAC+Vehicle组相比,TAC+OP-D组LVEF和FS显著升高(P<0.01和P<0.01);IVSd、LVPWd、心脏重量、HW/TL和心肌细胞CSA均显著减小(P<0.01、P<0.05、P<0.01、P<0.01和P<0.05);纤维化指数显著降低(P<0.001);心脏组织中Nppa、Nppb、Myh7、Tgfb1、Col1a1和Col1a3的转录水平均显著增加(P<0.05、P<0.001、P<0.05、P<0.001、P<0.001和P<0.01)。结论OP-D具有改善TAC小鼠心脏功能、减轻病理性心肌肥厚和心脏纤维化的作用。Objective To investigate the protective effect of ophiopogonin D(OP-D)on pathological myocardial hypertrophy induced by pressure overload in mice.Methods Transverse aortic constriction(TAC)was established to induce pressure overload pathological myocardial hypertrophy in mice.C57/BL6J mice were randomly divided into four groups using a random number generator,with eight mice in each group:Sham+Vehicle group,Sham+OP-D group,TAC+Vehicle group and TAC+OP-D.OP-D was administered via intraperitoneal injection.Cardiac function was evaluated by detecting left ventricular ejection fraction(LVEF)and short axis shortening rate(FS)using echocardiography.Pathological hypertrophy was assessed by measuring interventricular septum thickness(IVSd)and left ventricular posterior wall thickness(LPWd)of mice were measured by echocardiography,heart weight was weighed,the ratio of mouse heart weight to tibial length ratio(HW/TL),and calculating the average cardiomyocyte cross-sectional area(CSA)by wheat germ agglutinin staining.Cardiac fibrosis was detected by Masson staining.qPCR was used to detect the transcription levels of hypertrophy related molecules Nppa,Nppb,Myh7,and fibrosis related molecules Tgfb1,Col1a1,and Col1a3.Results Compared with the TAC+Vehicle group,after OP-D treatment,LVEF and FS in the TAC+OP-D group were significantly increased(P<0.01 and P<0.01);IVSd,LVPWd,heart weight,HW/TL,and the average CSA of myocardial cells were significantly reduced(P<0.01,P<0.05,P<0.01,P<0.01,and P<0.05,respectively);The fibrosis index significantly decreased(P<0.001);the transcription levels of Nppa,Nppb,Myh7,Tgfb1,Col1a1,and Col1a3 in cardiac tissue were all significantly increased(P<0.05,P<0.001,P<0.05,P<0.001,P<0.001,and P<0.01).Conclusion OP-D improved cardiac function,reduced myocardial hypertrophy,and alleviated fibrosis in TAC mice.
