摘  要:目的 研究金荞麦多糖(FCP)对人胃癌细胞SGC-7901的作用及其机制。方法 选择人胃癌细胞SGC-7901,常规培养及传代,设为阳性对照环磷酰胺(CTX)组、培养基阴性对照(空白)组及FCP 60、120、240 mg/L组。采用噻唑蓝(MTT)法观察细胞增殖,Hoechst33258染色法观察细胞凋亡,Western印迹和实时荧光定量-聚合酶链反应(PCR)法检测凋亡相关因子蛋白和mRNA表达。结果 FCP可明显抑制细胞增殖,与空白组比较,CTX组及FCP 60、120、240 mg/L组细胞增殖抑制率显著升高(P<0.05),FCP作用24、48、72、96 h的细胞抑制浓度(IC50)分别为(199.48±6.84)、(137.14±14.32)、(123.21±4.67)、(122.88±5.78)mg/L。与空白组比较,FCP可明显促进细胞凋亡,在荧光正置显微镜下可观察到典型细胞凋亡形态,如亮蓝色细胞核染色,细胞质浓缩和细胞核固缩等。与空白组比较,抑凋亡蛋白B细胞淋巴瘤(Bcl)-2 mRNA和蛋白表达显著下调(P<0.01);与空白组比较,60、120、240 mg/L FCP能明显促进细胞SGC-7901中胱天蛋白酶(Caspase)-3、8、9、Bcl-2相关X蛋白(Bax)、FasmRNA和蛋白及细胞色素(Cyt)C蛋白表达(P<0.01)。结论 FCP对SGC-7901细胞增殖和促凋亡作用突出,可能与提高Caspase-3、8、9、Bax蛋白表达、降低Bcl-2蛋白表达有关。Objective To study the effects of Fagopyrum Cymosum Polysaccharide(FCP)on human gastric cancer cells SGC-7901 and its mechanism.Methods Human gastric cancer SGC-7901 cells were selected for routine culture and passage,cells were divided into positive control cyclophosphamide(CTX)group,medium negative control(blank)group and FCP 60,120,240 mg/L groups.Thiazol blue(MTT)assay was used to observe cell proliferation,Hoechst33258 staining was used to observe cell apoptosis,Western blot and real-time fluorescence quantification-polymerase chain reaction(PCR)were used to detect the expression of apopto-sis-related factors protein and mRNA.Results Cell proliferation was significantly inhibited by FCP,compared with the blank group,the cell proliferation rate was significantly increased in CTX group and FCP 60,120,240 mg/L groups(P<0.05).50%inhibitory con-centration(IC50)for 24,48,72 and 96 h of FCP were respectively(199.48±6.84),(137.14±14.32),(123.21±4.67),(122.88±5.78)mg/L.Compared with the blank group,cell apoptosis and typical apoptotic morphology were significantly promoted by FCP,such as bright blue nuclear staining,cytoplasm concentration and nuclear pyrosis,could be observed under fluorescence micros-copy.Compared with blank group,the mRNA and protein expressions of anti-apoptotic protein B-cell lymphoma(Bcl)-2 were signifi-cantly down-regulated(P<0.01);compared with blank group,the expressions of Caspase-3,-8,-9,Bcl-2 associated X protein(Bax),Fas mRNA and protein and cytochrome(Cyt)C protein were significantly promoted by 60,120 and 240 mg/L FCP in SGC-7901 cells(P<0.01).Conclusions FCP had significant effects on proliferation and apoptosis of SGC-7901 cells and the mechanism may be re-lated to promoting the expressions of Caspase-3,-8,-9,Bax protein and inhibiting the expression of Bcl-2 protein.
