摘  要:目的探讨高糖高脂对人主动脉内皮细胞(HAECs)的氧化应激和凋亡的影响及通心络对其的干预作用。方法用高糖高脂建立细胞损伤模型,将HAECs随机分为空白对照组、高糖高脂组、通心络低、中、高浓度组。噻唑蓝(MTT)法检测细胞生存活性;基于组织的检测(TBA)法及羟胺法检测细胞上清液丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;流式细胞仪检测细胞内活性氧簇(ROS)水平和细胞凋亡率;Western印迹检测半胱氨酸天冬氨酸蛋白酶(Caspase)-3、B细胞淋巴瘤(Bcl)-2相关X蛋白(Bax)和Bcl-2蛋白表达。结果与空白对照组相比,高糖高脂组细胞生存活性、细胞上清液SOD活性明显降低,Bcl-2蛋白表达明显下调,细胞上清液MDA含量、ROS水平和细胞凋亡率明显升高,Caspase-3和Bax蛋白表达明显上调(P<0.01);与高糖高脂组相比,通心络中、高浓度组细胞生存活性、细胞上清液SOD活性明显升高,Bcl-2蛋白表达明显上调,细胞上清液MDA含量、细胞内ROS水平和细胞凋亡率明显降低,Caspase-3和Bax蛋白表达明显下调(P<0.05,P<0.01)。结论通心络能够改善高糖高脂诱导的HAECs功能损伤,其作用机制可能与减轻氧化应激反应和抑制凋亡有关。Objective To study the effects of high glucose and high lipid on oxidative stress and apoptosis of human aortic endothelial cells(HAECs)and the intervention effect of Tongxinluo.Methods The cell injury model was established by high glucose and high lipid,and HAECs were randomly divided into blank control group,high glucose and high lipid group,Tongxinluo low,medium and high concentration groups.Thiazolyl blue(MTT)assay was used to detect cell survival activity.The content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)in the supernatant were detected by thiobarbituric acid(TBA)method and hydroxylamine method.Intracellular reactive oxygen species(ROS)levels and apoptosis rate were detected by flow cytometry.The expressions of Caspase-3,B-cell lymphoma-2-associated X protein(Bax)and Bcl-2 protein were detected by Western blot.Results Compared with blank control group,the cell survival activity and the SOD activity in cell supernatant were significantly decreased,the Bcl-2 protein expression was significantly decreased,the MDA content in cell supernatant and the ROS level and the cell apoptosis rate were significantly increased,the Caspase-3 and the Bax protein expressions were significantly increased in high glucose and high lipid group(P<0.01).Compared with the high glucose and high lipid group,the cell survival activity and the SOD activity in cell supernatant were significantly increased,the expression of Bcl-2 protein was significantly increased,the content of MDA in the cell supernatant,the ROS level and the cell apoptosis rate were significantly decreased,and the protein expressions of Caspase-3 and Bax were significantly decreased in Tongxinluo medium and high concentration groups(P<0.05,P<0.01).Conclusions Tongxinluo could improve HAECs functional impairment induced by high glucose and high lipid,and its mechanism might be related to reducing oxidative stress and inhibiting apoptosis.
