摘 要:目的探究山奈酚(KPF)对阿尔茨海默病(AD)大鼠学习记忆功能及海马组织中极性蛋白分离缺陷(Par)3、Par6和非典型蛋白激酶(aPK)C表达的影响。方法将50只大鼠随机分为假手术组、模型组、阳性组(多奈哌齐)、KPF低、高剂量组,每组10只。Morris水迷宫实验检测学习记忆能力;苏木素-伊红(HE)染色检测海马组织的病理学形态;免疫荧光法检测海马组织β-淀粉样蛋白(Aβ)聚集情况;Western印迹检测海马组织中脑源性神经营养因子(BDNF)、酪氨酸蛋白激酶(Trk)B、Par3、Par6和aPKC蛋白表达。结果与假手术组相比,模型组逃避潜伏期和探索时间、Aβ聚集密度均明显升高,穿越平台次数、BDNF、TrkB、Par3、Par6和aPKC蛋白表达均明显降低(P<0.05);与模型组相比,阳性组和KPF各剂量组逃避潜伏期和探索时间、Aβ聚集密度均明显降低,穿越平台次数、BDNF、TrkB、Par3、Par6和aPKC蛋白表达均明显升高(P<0.05)。结论KPF可以增强AD大鼠的学习记忆能力,促进海马组织中极性蛋白Par3、Par6和aPKC表达。Objective To investigate the influences of kaempferol(KPF)on learning and memory function and the expressions of polar protein separation defect(Par)3,Par6 and atypical protein kinase(aPK)C in hippocampus of Alzheimer's disease(AD)rats.Methods Fifty rats were randomly divided into sham operation group,model group,positive group(donepezil),KPF low and high dose groups,with 10 rats in each group.Morris water maze test was used to test the learning and memory ability;hematoxylin-eosin(HE)staining was used to detect the pathological morphology of hippocampus;the aggregation ofβ-amyloid(Aβ)in hippocampus was detected by immunofluorescence method;Western blot was used to detect the protein expressions of brain-derived neurotrophic factor(BDNF),tyrosine protein kinase(Trk)B,Par3,Par6 and aPKC in hippocampus.Results Compared with the sham operation group,the escape latency,exploration time and Aβaggregation density were significantly increased in the model group,and the number of platform crossing,the protein expressions of BDNF,TrkB,Par3,Par6 and aPKC were significantly decreased in model group(P<0.05);compared with the model group,the escape latency,exploration time and Aβaggregation density of positive group and KPF dosage groups were significantly decreased,while the number of platform crossing,the protein expressions of BDNF,TrkB,Par3,Par6 and aPKC were significantly increased(P<0.05).Conclusions KPF could enhance the learning and memory ability of AD rats and promote the expressions of polar protein Par3,Par6 and aPKC in hippocampus.
