摘 要:试验旨在探究转ATP合酶A亚基基因(atpA)、ATP合酶B亚基基因(atpB)拟南芥在逆境(干旱、盐胁迫、冷胁迫)下的生长发育、光合作用及能量代谢响应。采用酶标法测定超氧化物歧化酶(SOD)、腺苷三磷酸酶(ATPase)、过氧化物酶(POD)、过氧化氢酶(CAT)活性,以及腺嘌呤核苷三磷酸(ATP)含量,系统评估了不同PEG-6000浓度(0、15%、20%、25%、30%)、盐浓度(0、100、200、250、300 mmol/L)及冷胁迫(4℃)下转atpA、atpB基因拟南芥的生理变化。结果显示:随着冷胁迫时间的增加,转atpA基因拟南芥ATP含量呈先增加后降低的趋势,转atpB基因拟南芥ATP含量先降低后保持平稳。在200、250 mmol/L NaCl胁迫下,胁迫3 h转atpA基因拟南芥的ATP含量均最高,在250、300 mmol/L NaCl胁迫下,随着胁迫时间的增加,转atpB基因拟南芥ATPase的活性呈先上升后逐渐下降的趋势。在15%、20%、25%PEG-6000胁迫下,转atpA基因拟南芥的ATPase活性在3~12 h逐渐下降,12 h后逐渐上升。研究表明,转atpA、atpB基因拟南芥在逆境下的生理响应存在明显差异,可能与基因改造后的特定功能及逆境适应机制有关,可为转基因作物的抗逆性改良提供参考。The aim of this experiment was to investigate the growth,development,photosynthesis and energy metabolism response of transgenic atpA and atpB Arabidopsis genes under stress(drought,salt stress,cold stress).The activities of superoxide dismutase(SOD),adenosine triphosphatase(ATPase),peroxidase(POD),catalase(CAT),and the content of adenine riboside triphosphate(ATP)were determined by enzymolysis method.The physiological changes of transgenic atpA and atpB genes Arabidopsis were systematically evaluated under different PEG-6000 concentrations(0,15%,20%,25%,30%),salt concentrations(0,100,200,250,300 mmol/L)and cold stress(4℃).The results showed that with the increase of cold stress time,the ATP content of transgenic atpA Arabidopsis first increased and then decreased,and the ATP content of transgenic atpB Arabidopsis first decreased and then remained stable.Under 200 and 250 mmol/L NaCl stress,the ATP content in transgenic atpA Arabidopsis was the highest after 3 hours of stress.Under 250 and 300 mmol/L NaCl stress,the activity of ATPase in transgenic atpB Arabidopsis showed a trend of increasing first and then gradually decreasing with the extension of stress time.Under 15%,20%,and 25%PEG-6000 stress,the ATPase activity in transgenic atpA Arabidopsis gradually decreased from 3 to 12 hours and then gradually increased after 12 hours.The results show that there are significant differences in the physiological response of transgenic atpA and atpB gene Arabidopsis under stress,which may be related to the specific function and stress adaptation mechanism after gene modification.It can provide reference for the improvement of stress resistance of transgenic crops.
