制何首乌对糖尿病性白内障大鼠晶状体上皮细胞的影响

摘  要:目的 研究制何首乌对糖尿病性白内障(DC)大鼠晶状体上皮细胞的保护作用。方法 随机选取60只健康SD雄性大鼠通过腹腔一次性注射链脲佐菌素诱导糖尿病模型,造模成功后随机分为对照(NC)组、DC模型(DC)组、制何首乌低剂量(低RPM)组和制何首乌高剂量(高RPM)组,每组15只;裂隙灯监测大鼠白内障的形成情况;苏木素-伊红(HE)染色观察晶状体的病理学变化;TUNEL染色检测晶状体上皮细胞凋亡;RT-聚合酶链反应(PCR)和Western印迹检测B细胞淋巴瘤(Bcl)-2相关X蛋白(Bax)、Bcl-2和胱天蛋白酶(Caspase)-3 mRNA和蛋白表达;酶联免疫吸附试验(ELISA)检测晶状体中白介素(IL)-1β和IL-6含量。结果 与NC组相比,DC组晶状体混浊程度明显加重,甚至完全混浊,糖尿病大鼠出现白内障;相反,低RPM和高RPM组晶状体混浊程度得到缓解。HE染色显示,NC组晶状体上皮细胞形态规则、排列整齐,DC组晶状体上皮细胞形态大小不一,排列紊乱;制何首乌干预后晶状体上皮细胞损伤程度得到明显缓解。与NC组比较,DC组细胞凋亡率、Bax、Caspase-3 mRNA及蛋白水平、IL-6及IL-1β含量明显升高,Bcl-2 mRNA及蛋白水平明显降低(P<0.05);与DC组比较,高RPM组细胞凋亡率、Bax、Caspase-3 mRNA及蛋白水平、IL-6及IL-1β含量明显降低,Bcl-2 mRNA及蛋白水平明显升高(P<0.05)。结论 糖尿病引起的白内障与Bax、Caspase-3表达增加、Bcl-2的抑制及炎症因子的激活有关。制何首乌可通过上调Bcl-2表达,降低Bax和Caspase-3表达和炎症因子的水平从而延缓白内障发生。Objective To study the protective effect of Prepared Radix Polygoni Multiflori(RPM)on the epithelial cells of lens in diabetic cataract(DC)rats.Methods 60 healthy SD male rats were randomly selected to induce diabetes model by one-time intraperitoneal injection of streptozotocin.After successful modeling,rats were randomly divided into control(NC)group,DC model(DC)group,low dose RPM(low RPM)group and high dose RPM(high RPM)group,with 15 rats in each group;slit lamp was used to monitor cataract formation in rats;hematoxylin-eosin(HE)staining was used to observe the pathological changes of lens;TUNEL staining was used to detect apoptosis of lens epithelial cells;RT-polymerase chain reaction(PCR) and Western blot were used to detect themRNA and protein expressions of B-cell lymphoma(Bcl)-2 associated X protein(Bax),Bcl-2 and Caspase-3;enzyme linked immu-nosorbent assay (ELISA) was used to detect the contents of interleukin (IL)-1β and IL-6 in the lens. Results Compared with theNC group, the turbidity of the lens in the DC group was significantly increased, even completely turbid, and cataracts appeared in dia-betic rats;on the contrary, the turbidity of the lens was alleviated in low RPM and high RPM groups. HE staining showed that the mor-phology of the lens epithelial cells in the NC group was regular and well-arranged, the morphology of the lens epithelial cells in the DCgroup was varied, and the arrangement of the cells was disordered;the extent of the damage to the lens epithelium was significantly al-leviated after the intervention of RPM. Compared with the NC group, the apoptosis rate, Bax, Caspase-3 mRNA and protein levels,IL-6 and IL-1β contents were significantly increased, and Bcl-2 mRNA and protein levels were significantly decreased in the DC group(P<0. 05);compared with the DC group,the apoptotic rate, Bax, Caspase-3 mRNA and protein levels, IL-6 and IL-1β contents weresignificantly decreased, Bcl-2 mRNA and protein levels were significantly increased in the high RPM group(P<0. 05).
