摘 要:目的 探讨电针对老年大鼠骨质疏松及组织蛋白酶K、Ⅰ型胶原表达的影响。方法 选取8只6月龄SD雄性大鼠为青年组,16只24月龄SD雄性大鼠随机分为老年组和电针组,每组8只。电针组接受电针干预,电针穴位:“肾俞”“血海”“太溪”“阳陵泉”。电流强度1 mA,疏密波,频率3 Hz/15 Hz,每周5 d, 1次/d,连续干预8 w,其余2组不接受干预。8 w后取材,酶联免疫吸附试验(ELISA)检测外周血骨代谢标志物Ⅰ型前胶原氨基末端肽(PⅠNP)、Ⅰ型胶原交联氨基末端肽(NTX)水平,双能X射线检测左侧胫骨和L5椎体的骨密度,显微CT检测右胫骨近端和L4椎体的骨微结构,Western印迹、实时荧光定量-聚合酶链反应(RT-qPCR)检测股骨组织组织蛋白酶K、Ⅰ型胶原蛋白及mRNA表达水平。结果 与青年组相比,老年组NTX水平升高,PⅠNP水平降低,骨密度、骨体积分数、骨小梁数量均降低,骨小梁间距增大,组织蛋白酶K蛋白及mRNA表达升高,Ⅰ型胶原蛋白及mRNA表达降低,差异有统计学意义(P<0.05)。与老年组相比,电针组NTX水平降低,PⅠNP水平升高,骨密度、骨体积分数、骨小梁数量升高,骨小梁间距降低,组织蛋白酶K蛋白及mRNA表达降低,Ⅰ型胶原蛋白及mRNA表达升高,差异有统计学意义(P<0.05)。结论 电针通过下调组织蛋白酶K的表达从而抑制Ⅰ型胶原蛋白的降解,达到治疗骨质疏松的作用。Objective To investigate the effect of electroacupuncture on osteoporosis and the expressions of cathepsin K and collagen-Ⅰin aged rats.Methods A total of eight 6-month-old SD male rats were selected as the young group,and sixteen 24-month-old SD male rats were randomly divided into elderly group and electroacupuncture group,with eight rats in each group.The electroacu-puncture group were given electroacupuncture intervention using the following acupoints:"Shenshu""Blood Sea""Taixi"and"Yan-glingquan". The current intensity was set at 1 mA, using a density wave with a frequency of 3 Hz/15 Hz. The intervention was admin-istered five days a week, once per day, for a continuous period of eight weeks. Meanwhile, the other two groups were not given any in-tervention. After 8 w, samples were collected and analyzed using enzyme-linked immunosorbent assay (ELISA) to detect the levels ofperipheral blood bone metabolic markers of type Ⅰ procollagen (PⅠNP) and type Ⅰ collagen cross linked N telopeptides (NTX) inperipheral blood. The bone mineral density of the left tibia and L5 vertebra was assessed using dual-energy X-ray, while the bone mi-crostructure of the right proximal tibia and L4 vertebra was examined using micro-CT. Furthermore, the mRNA and protein expressionlevels of cathepsin K and collagen-Ⅰ in femur tissue were determined using Western blotting and real-time fluorescence quantitative re-verse transcription polymerase chain reaction (RT-qPCR). Results Compared with the young group, NTX level was increased, PⅠNP level was decreased, bone density, bone volume fraction, trabecular thickness and trabecular number were all decreased, trabecu-lar bone spacing was increased, the mRNA and protein expressions of cathepsin K were increased, the mRNA and protein expressionsof collagen-Ⅰ were decreased, the difference was statistically significant in elderly group(P<0. 05).
